目的 探讨半乳凝素1( galectin-1)在口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)中的表达及临床意义,旨作为OSCC早期诊断的检测指标。方法分别采用实时定量聚合酶链反应(PCR)、蛋白质印迹法和免疫组化方法检测口腔黏膜上皮体外癌变模型、OSCC细胞系及30例原发OSCC标本中的半乳凝素1 mRNA及蛋白质的表达。结果半乳凝素1在永生化细胞系(humanimmortalized oral epithelia cell,HIOEC)中mRNA和蛋白相对表达量平均为0.071±0.023和0.118±0.046,与永生化细胞系HIOEC相比,半乳凝素1在所有检测的体外癌变模型和OSCC细胞系中mRNA和蛋白相对表达水平均升高,分别为0.141±0.049和0.504±0.33(P<0.01)。30例OSCC组织中半乳凝素1 mRNA和蛋白相对表达量(0.059±0.034、1.5±0.68)均显著高于癌旁组织(0.029±0.012、0.4±0.56) (P<O.01)。结论半乳凝素1在口腔黏膜上皮癌变过程中表达上调可能与OSCC的发生、发展有关,可作为OSCC早期诊断的检测指标。

목적 탐토반유응소1( galectin-1)재구강린상세포암(oral squamous cell carcinoma,OSCC)중적표체급림상의의,지작위OSCC조기진단적검측지표。방법분별채용실시정량취합매련반응(PCR)、단백질인적법화면역조화방법검측구강점막상피체외암변모형、OSCC세포계급30례원발OSCC표본중적반유응소1 mRNA급단백질적표체。결과반유응소1재영생화세포계(humanimmortalized oral epithelia cell,HIOEC)중mRNA화단백상대표체량평균위0.071±0.023화0.118±0.046,여영생화세포계HIOEC상비,반유응소1재소유검측적체외암변모형화OSCC세포계중mRNA화단백상대표체수평균승고,분별위0.141±0.049화0.504±0.33(P<0.01)。30례OSCC조직중반유응소1 mRNA화단백상대표체량(0.059±0.034、1.5±0.68)균현저고우암방조직(0.029±0.012、0.4±0.56) (P<O.01)。결론반유응소1재구강점막상피암변과정중표체상조가능여OSCC적발생、발전유관,가작위OSCC조기진단적검측지표。
Objective To investigate the expression of galectin-1 in oral squarnous cell carcinoma (OSCC) and its clinical significance. Methods Detection of the mRNA and protein expression of galectin-1 in the in vitro cellular carcinogenesis model of OSCC, OSCC cell lines and tissue specimens from 30 primary OSCC patients were performed using real-time polymerase chain reaction (PCR), Western blotting and immunohistochemistry, respectively. Results The value of galectin-1 mRNA and protein level in human immortalized oral epithelia cell (HIOEC) cell was 0. 071 ±0.023,0.118 ±0.046, Compared with the HIOEC, galectin-1 mRNA level and protein expression were increased significantly in all the cell lines (0. 141 ±0. 049,0. 504 ±0. 33) ( P <0. 01 ). The levels of mRNA and protein expression of galectin-1 were significantly higher in the cancerous tissue(0. 059 ±0. 034,1.5 ±0. 68) than in the normal adjacent tissues (0. 029 ±0. 012,0. 4 ±0. 56 ) ( P <0. 01 ). Conclusions The expression of galectin-1 gene up-regulated in carcinogenesis process of OSCC significantly may be related to the tumorigenesis and development of OSCC,which illustrates its potential clinical application as tumor marker for early diagnosis.
