利用原代培养后纯化的奶牛乳腺上皮细胞,以Matrigel为基质胶原,进行奶牛乳腺上皮细胞的三维培养,细胞接种浓度分别为1×104、1×105和1×106个细胞/mL。经过16d培养后,用DAPI对细胞染色,利用荧光显微镜观察细胞生长状况。结果表明,当细胞接种浓度为1×104~1×105个细胞/mL时,细胞形成了团块状结构,出现了类似腔状的腺泡形态。
利用原代培養後純化的奶牛乳腺上皮細胞,以Matrigel為基質膠原,進行奶牛乳腺上皮細胞的三維培養,細胞接種濃度分彆為1×104、1×105和1×106箇細胞/mL。經過16d培養後,用DAPI對細胞染色,利用熒光顯微鏡觀察細胞生長狀況。結果錶明,噹細胞接種濃度為1×104~1×105箇細胞/mL時,細胞形成瞭糰塊狀結構,齣現瞭類似腔狀的腺泡形態。
이용원대배양후순화적내우유선상피세포,이Matrigel위기질효원,진행내우유선상피세포적삼유배양,세포접충농도분별위1×104、1×105화1×106개세포/mL。경과16d배양후,용DAPI대세포염색,이용형광현미경관찰세포생장상황。결과표명,당세포접충농도위1×104~1×105개세포/mL시,세포형성료단괴상결구,출현료유사강상적선포형태。
The three-dimensional cultures of bovine mammary epithelial cells were conducted with Matrigel.The cell inoculum density was 1×104,1×105 and 1×106 cells/mL,respectively.After 16 days culture,the cells were stained with DAPI and the cell morphology were detected by fluorescence microscope.The results suggested that when the density was 1×104 and 1×105 cell/mL the cells were in a cluster,and acinar structures were formed.
