目的 观察大蒜素对胃癌细胞株及移植瘤的作用,初步探讨抗癌机制。方法 采用MTT法观察不同质量浓度及作用不同时间大蒜素对胃癌SGC-7901及MGC-803细胞的影响;流式细胞术分析100mg*L-1大蒜素分别作用4h和8h后两种肿瘤细胞增殖周期的变化;荷胃癌SGC-7901裸鼠模型随机分成4组,每组4只,分别于荷瘤7d和14d用20g/L的大蒜素0.1mL和PBS0.1mL对瘤体进行局部注射治疗,每隔2d一次,共3次,观察瘤体增殖及裸鼠生存状况的变化。结果 不同质量浓度梯度大蒜素对两种肿瘤细胞皆有一定的杀伤,大蒜素质量浓度为800mg.L-1及1000mg.L-1时,对SGC-7901细胞的抑制率分别为65.5%,68.6%(P<0.01,vs5-FU)。大蒜素对MGC-803细胞的抑制作用较明显,质量浓度为400mg.L-1,800mg.L-1及1000mg.L-1时,抑制率分别为67.5%,75.3%和79.5%(P<0.01,vs5-FU)。100mg.L-1大蒜素连续作用3d,对SGC-7901及MGC-803的抑制率分别可达85.2%,96.7%(P<0.01,vs5-FU)。大蒜素作用后SGC-7901及MGC-803细胞S期比例有增高趋势。荷瘤7d局部注射治疗组有3只裸鼠瘤体消失,余下1只瘤体组织切片可见大量淋巴细胞聚集,肿瘤血管较少。荷瘤14d局部注射治疗组4只荷瘤裸鼠的瘤体生长延缓,生存状态较好,瘤体组织切片中可见大片肿瘤组织坏死。结论 大蒜素可能通过直接杀伤胃癌细胞,提高机体免疫等方面的综合作用抑制胃癌细胞生长。

Objective To study the anticarcinogen mechanism of garlicin afterobservation of the effect of garlicin on gastric cancer cells.Methods Cultural cell lines of gastric cancer SGC-7901 and MGC-803 were used as the target cells to observe the effect of garlicin of different concertritions with different culture time on these cell lines by MTT assay.Two kinds of gastrical cancer cells with 10% bovine serum and 100mg.L-1 garlicin were cultured,and their changs of cell cycles after 4 and 8 hours were measured in vitro.In vivo study,garlicin were injected locally into the inplanted neoplasm of Balb/c nu/nu mice to observe the changs of the neoplasms and the living state after 7 and 14 days respectively when the nude mice tumor xenograft model were made.Results Garlicin had cytotoxic effect on these two kinds of gastrical cancer cells.When the mass concentritions of garlicin were 800mg.L-1 and 1000mg.L-1,the inhibiting rates of on SGC-7901 cells were 65.5% and 68.6% respectively(P<0.01,vs5-FU).When the mass concentritions of garlicin were 400mg.L-1,800mg.L-1 and 1000mg.L-1,the inhibiting rates on MGC-803 cells were 67.5%,75.3% and 79.5 respectively(P<0.01,vs5-FU).Culturing two kinds of gastrical cancer cells with bovine serum and 100mg.L-1 garlicin for 3 days,the inhibiting rates could come up to 85.2% and 96.7% respectively.Culturing two kinds of gastrical cancer cells with bovine serum and 100mg.L-1garlicin for 4 and 8 hours,the S phase of the cell cycles rose,and that of the MGC-803 cells rose more obviously.Injecting garlicin locally into the neoplasms in the seventh day when the nude mice tuor xenograft models were made,the neoplasm subsided in three nude mice,the neoplasm of one nude mice became very small.There were plenty of lymphocytes in the neoplasms,and the vasalium in neoplasm was scarce.Garlicin depressed the progress of the neoplasms when the nude mice tumor xenograft models were made by injecting locally in the fourteenth days,whose living state were better than that of controls.There were larges of areas of necrosis in the neoplasms.Conclusion Garlicin has depressive effection possiblely by killing cancer cells directly and improving the function of immunity.
