为研究昆山接骨片对体外成骨细胞骨形成功能的影响,探讨昆山接骨片促进骨折愈合的机理,分别用MTT法观察了含昆山接骨片Ⅰ号和昆山接骨片Ⅱ号的药物血清对成骨细胞增殖功能的影响,用PNPP法观察昆山接骨片Ⅰ号对成骨细胞碱性磷酸酶比活性的影响,以及用矿化结节计数法观察了昆山接骨片Ⅰ号对成骨细胞矿化功能的影响。成骨细胞增殖率测定结果显示,昆山接骨片Ⅰ号组为0.092±0.016,与对照组比较有显著性差异(P<0.05);而Ⅱ号组为0.230±0.026,与对照组比较无差异。ALP比活性测定结果显示,昆山接骨片Ⅰ号组为0.034±0.021,对照组为0.026±0.008,两组比较有显著性差异(P<0.05))。矿化结节计数结果提示,昆山接骨片Ⅰ号组为25.833±7.782,对照组为13.333±6.153,两组比较有显著性差异(P<0.05)。综合研究结果表明,以活血化瘀药物为主制成的昆山接骨片Ⅰ号具有促进成骨细胞的增殖、分化与矿化、成骨细胞的骨形成功能,从而发挥其促进骨折愈合的作用,单纯用螃蟹粉制成的昆山接骨片Ⅱ号则无此作用。

위연구곤산접골편대체외성골세포골형성공능적영향,탐토곤산접골편촉진골절유합적궤리,분별용MTT법관찰료함곤산접골편Ⅰ호화곤산접골편Ⅱ호적약물혈청대성골세포증식공능적영향,용PNPP법관찰곤산접골편Ⅰ호대성골세포감성린산매비활성적영향,이급용광화결절계수법관찰료곤산접골편Ⅰ호대성골세포광화공능적영향。성골세포증식솔측정결과현시,곤산접골편Ⅰ호조위0.092±0.016,여대조조비교유현저성차이(P<0.05);이Ⅱ호조위0.230±0.026,여대조조비교무차이。ALP비활성측정결과현시,곤산접골편Ⅰ호조위0.034±0.021,대조조위0.026±0.008,량조비교유현저성차이(P<0.05))。광화결절계수결과제시,곤산접골편Ⅰ호조위25.833±7.782,대조조위13.333±6.153,량조비교유현저성차이(P<0.05)。종합연구결과표명,이활혈화어약물위주제성적곤산접골편Ⅰ호구유촉진성골세포적증식、분화여광화、성골세포적골형성공능,종이발휘기촉진골절유합적작용,단순용방해분제성적곤산접골편Ⅱ호칙무차작용。
Objective:To study the effect of “Kunshan Jiegu” Tablet(KJT) onthe bone formation of osteoblasts.Methods:MTT method was used to observe the effects of blood serums of KJT-I and KJT-II on the proliferation of osteoblasts;PNPP method was used to evaluate the effect of KJT-I on the specific activity of alkaline phosphatase of osteoblasts;The counting method of mineralization nodules was used to observe the effect of KJT-I on osteoblasts' mineralization.Results:The effect of KJT-I group on the proliferation of osteoblasts is significantly different from that of the control group,while the result of KJT-II is on the contrary.The effect of KJT-I group on the activities of osteoblasts and mineralization differs greatly from that of the control group.Conclusion:KJT-I could increase proliferation,differentiation and minralization of osteoblasts,thus could inprove the fracture healing.
