本研究采用醇沉淀法从350g玉米花粉提取得粗提复合物38.3g。经用蒽酮反应、α萘酚反应、间苯二酚反应、碘化反应及高效液相色谱法鉴定,证实为多糖。用上述多糖作为免疫增强剂按剂量50、100、200mg分别与猪瘟弱毒细胞冻干苗共同免疫猪,测定其免疫效果。结果表明,各剂量组的IHA抗体效价均高于对照组;T细胞活性E花环试验测得各剂量的活性E花环百分率分别为21.84%、25.46%、21.92%,均高于对照组15.91%;T淋巴细胞转化试验显示,各试验组的T淋巴细胞转化率分别为55.32%、56.39%、54.27%,均高于对照组47%,上述三种方法测定的结果均显示显著差异。对T淋巴细胞亚群的测定结果为,各试验组的CD3、CD4、CD8T淋巴细胞的测定平均值均明显高于对照组,但NK细胞的变化不显著。根据试验结果选取100mg剂量组在两个猪场进行田间扩大试验,测得的IHA抗体效价亦高于对照组。研究结果表明,玉米花粉多糖既可增强体液免疫,又能增强细胞免疫。

본연구채용순침정법종350g옥미화분제취득조제복합물38.3g。경용은동반응、α내분반응、간분이분반응、전화반응급고효액상색보법감정,증실위다당。용상술다당작위면역증강제안제량50、100、200mg분별여저온약독세포동간묘공동면역저,측정기면역효과。결과표명,각제량조적IHA항체효개균고우대조조;T세포활성E화배시험측득각제량적활성E화배백분솔분별위21.84%、25.46%、21.92%,균고우대조조15.91%;T림파세포전화시험현시,각시험조적T림파세포전화솔분별위55.32%、56.39%、54.27%,균고우대조조47%,상술삼충방법측정적결과균현시현저차이。대T림파세포아군적측정결과위,각시험조적CD3、CD4、CD8T림파세포적측정평균치균명현고우대조조,단NK세포적변화불현저。근거시험결과선취100mg제량조재량개저장진행전간확대시험,측득적IHA항체효개역고우대조조。연구결과표명,옥미화분다당기가증강체액면역,우능증강세포면역。
In this study we had obtainded 38.8g of crude extract from cornpollen using alcohol precipitation method. The crude extract was identified as polys accharides after detection by anthraone,molish, seliwanoff, iodide reactions and high-performance liquid-phase chromatograph measure. The corn pollen polysaccharides(CPP) was used as immunol ogical enhancer and its effect was detected by using 50, 100 and 200 mg of CPP m ixing with hog cholera lapinized virus (HCLV). As result, IHA antibody titers of three groups using CPP were higher than that of control. In the T lymphocyte ac tive-Erosette test, it showed average rate 21.84%, 25.46% and 21.92% of E-rosette in three groups and all groups were higher than 15.91% of control. The Tlymp hocyte transformation rate of three experimental groups were 55.32%, 56.39% and 54.27 % ,respectively, and three groups were higher than 47% of control. The results of three methods demonstrated that CPP could promote immune effect when it was mi xed with HCLV vaccine. T lymphocyte subgroup assay was also carried out by count ing CD3, CD4 and CD8.T lymphocyte and their average value were higher than that of control. However, NK cells of three experimental groups did not show signific antly changes contrast with that of control. According to results above,100 mg CPP was selected to do field trial, and the IHA antibody level was also showed hig her than that of control. The results in this experiment demonstrated that CPP could be used as an immunological enhancer to promote humoral and cellular-immunity.
