目的:观察化痰方(二陈汤)对高脂饮食大鼠血脂及胰岛素抵抗的影响。方法:30只健康成年雄性大鼠按照随机数字表法分为正常组、高脂组和化痰组各10只,正常组给予正常饲料喂养,高脂组和化痰组给予高脂饲料喂养14周,化痰组第11周起给予二陈汤灌胃4周。在第14周末进行葡萄糖耐量实验(OGTT)和胰岛素耐量实验(ITT),14周后处死大鼠,检测并比较各组血脂TC、TG的含量及第14周各组大鼠OGTT结果和ITT结果。结果:与正常组比较,高脂组大鼠血清中的TC和TG含量均明显升高(P<0.01);与高脂组比较,化痰组大鼠血清中的TG含量明显降低(P<0.01),TC有所降低,差异也有统计学意义(P<0.05)。高脂组的空腹血糖明显高于正常组(P<0.05),而化痰组空腹血糖与正常组的比较差异无统计学意义(P>0.05);糖负荷后,高脂组的各个时间点的血糖均明显高于正常组(P<0.05或P<0.01),而化痰组除了糖负荷后120 min的血糖明显高于正常组外(P<0.01),其余时间点与正常组的比较差异均无统计学意义(P>0.05)。腹腔注射胰岛素后,化痰组各时间点血糖下降幅度均明显高于高脂组,30 min时高脂组血糖降低18%,而化痰组大鼠降低26%。结论:二陈汤能够降低高脂饮食大鼠血脂的含量,改善胰岛素抵抗,值得在临床上推广使用。

목적:관찰화담방(이진탕)대고지음식대서혈지급이도소저항적영향。방법:30지건강성년웅성대서안조수궤수자표법분위정상조、고지조화화담조각10지,정상조급여정상사료위양,고지조화화담조급여고지사료위양14주,화담조제11주기급여이진탕관위4주。재제14주말진행포도당내량실험(OGTT)화이도소내량실험(ITT),14주후처사대서,검측병비교각조혈지TC、TG적함량급제14주각조대서OGTT결과화ITT결과。결과:여정상조비교,고지조대서혈청중적TC화TG함량균명현승고(P<0.01);여고지조비교,화담조대서혈청중적TG함량명현강저(P<0.01),TC유소강저,차이야유통계학의의(P<0.05)。고지조적공복혈당명현고우정상조(P<0.05),이화담조공복혈당여정상조적비교차이무통계학의의(P>0.05);당부하후,고지조적각개시간점적혈당균명현고우정상조(P<0.05혹P<0.01),이화담조제료당부하후120 min적혈당명현고우정상조외(P<0.01),기여시간점여정상조적비교차이균무통계학의의(P>0.05)。복강주사이도소후,화담조각시간점혈당하강폭도균명현고우고지조,30 min시고지조혈당강저18%,이화담조대서강저26%。결론:이진탕능구강저고지음식대서혈지적함량,개선이도소저항,치득재림상상추엄사용。
Objective:To observe the effect of Erchen decoction on blood fat and insulin resistance induced by high-fat diet in rats.Method:Thirty adult male rats were randomly divided into three groups:the normal group,the high-fat group and the phlegm group.The normal group was fed with normal diets,the high-fat group and phlegm group were fed with high fat diets for 14 weeks.The phlegm group group was given with Erchen decoction for 4 weeks starting from eleventh weeks.At the end of the fourteenth week,oral glucose tolerance test and insulin tolerance test were performed for estimating insulin sensitivity.All rats were sacrificed at the 14 weeks,the concentration of TC and TG in serum,OGTT and ITT results of all the groups at fourteenth weeks were determined and compared.Result:Compared with the normal group,TC and TG in serum of the high-fat group increased significantly(P<0.01).Compared with the high-fat group, the TG content in serum of the phlegm group significantly decreased(P<0.01),TC decreased,and the difference was also statistically significant(P<0.05).The fasting blood glucose of high-fat group was significantly higher than that of normal group,but there was no significant difference between the phlegm group and the normal group(P>0.05).After the glucose load,each time point of blood sugar in high-fat group were significantly higher than those in normal group (P<0.05 or P<0.01).But the phlegm group except after glucose loading 120 min glucose was significantly higher than normal group(P<0.01),the rest of the time points compared with normal group had no statistical significance(P>0.05). After intraperitoneal injection of insulin,phlegm group at each time point of blood glucose decreased were significantly higher than the high lipid group,blood glucose of high-fat group at 30 min decreased by 18%,while the phlegm group decreased by 26%.Conclusion:The Erchen decoction can reduce lipid content in rats fed with high fat diet,improve insulin resistance,it is worth in clinical use.
