目的探讨三氧化二砷诱导表达带3蛋白膜段结构域K562细胞凋亡的作用.方法将纯化的表达载体pYD1-mdb3转染K562细胞, Western Blot检定有带3蛋白膜段结构域的表达;通过光学显微镜、DNA电泳和线粒体膜电位的测定,观察三氧化二砷对表达带3蛋白膜段结构域的K562细胞凋亡的影响.结果表达带3蛋白膜段结构域的K562细胞的上述凋亡特征均比未表达带3蛋白的K562细胞明显.结论表达带3蛋白膜段结构域的K562细胞对三氧化二砷更敏感.

Objective To investigate the function of band 3 membrane domain during arsenic trioxide (As2O3) induced apoptosis of K562 cells.Methods The purified expression vector pYD1-mdb3 was transfected into K562 cells. The expressed band 3 membrane domain was identified by Western blot. The effect of As2O3 on K562 cells which expressed band 3 membrane domain was observed with light microscope, DNA electrophoresis, and mitochondrial membrane potential(MMP)assay. Results As2O3 induced more significant changes of apoptosis in K562 cells which expressed membrane domain of band 3 protein. Conclusion The K562 cells expressed band 3 membrane domain are more susceptible to As2O3.
