将枯草芽孢杆菌(Bacillus subulis)基因组DNA按5、20和80 μg/kg剂量体腔注射中华绒螯蟹(Eriocheir sinensis)(约0.1 mL/只),设注射等体积TE缓冲液为对照组,分别在第1、3、5、7天采样,测定该细菌DNA对中华绒螯蟹的免疫刺激作用.结果显示:注射上述3种剂量的枯草芽孢杆菌基因组DNA均能不同程度地提高中华绒螫蟹的THC、血细胞呼吸爆发活性和血清PO、ACP活性,试验组蟹的上述免疫指标的活性显著高于对照组(P<0.05),并以20 μg/kg体重剂量的刺激效果最好;但试验剂量的细菌DNA对血清ALP活性无显著影响(P>0.05).

장고초아포간균(Bacillus subulis)기인조DNA안5、20화80 μg/kg제량체강주사중화융오해(Eriocheir sinensis)(약0.1 mL/지),설주사등체적TE완충액위대조조,분별재제1、3、5、7천채양,측정해세균DNA대중화융오해적면역자격작용.결과현시:주사상술3충제량적고초아포간균기인조DNA균능불동정도지제고중화융석해적THC、혈세포호흡폭발활성화혈청PO、ACP활성,시험조해적상술면역지표적활성현저고우대조조(P<0.05),병이20 μg/kg체중제량적자격효과최호;단시험제량적세균DNA대혈청ALP활성무현저영향(P>0.05).
The immtmostimulatory effects of genomic DNA of Bacillus subtilis on Chinese mitten crab (Eriocheir sinensis) were studied when the crabs were injected through coelom by dose of 5,20 and 80 μg/kg body weight (about 0.1 mL/crab) individually,and one group injected with 0.1 mL TE buffer was set as a control.The major immune parameters of E.sinensis were examined at 1,3,5 and 7 days after administration of bacterial DNA or TE buffer.The results showed that three doses of bacterial DNA could enhance THC,respiratory burst of haemocytes,PO and ACP activities in serum of E.sinensis to different extent.The activities of the above immune parameters of crabs in three test groups were significantly higher than those of the control group at most of the sampling time points (P<0.05).The immunostimulatory effects of bacterial DNA on E.sinensis by injection at 20 μg/kg body weight were the best.And no significant difference on the ALP activities in serum of E.sinensis was detected after the administration of bacterial DNA (P>0.05).
