以酶水解液为发酵基质,采用Plackett-Burman设计法对影响丙酮丁醇梭菌(Clostridium acetobutylicum CICC 8008)厌氧发酵生产丁醇的15个因子进行筛选.结果表明,影响该菌的厌氧发酵的主要影响因子为CaCO_3.在该基础上采用响应曲面法对CaCO_3浓度、温度、时间3个因子的影响进行研究,得出多元二次回归方程拟合3种因素与丁醇产量间的函数关系,并根据优化结果与试验验证,当CaCO_3浓度为5.04 g/L,温度为35 ℃,时间为70 h,丁醇发酵从初始发酵培养条件下的5.15 g/L提高到6.20 g/L,产量提高了20%.

With enzymatic hydrolysis as the fermentation substrate, Plackett-Burman design method was adopted to screen fifteen influencing factors for butanol production by Clostridium acetobutylicum CICC 8008. The results showed the key factor was CaCO_3. Based on these results, response surface method was used for further study on the effects of CaCO_3, temperature and reaction time in order to find out the optimum concentration levels. The concentration of butanol could reach 6.20 g/L, compared to initial 5.15 g/L, and the yield was enhanced by 20% under the conditions of CaCO_3 concentration 5.04 g/L, temperature 35 ℃, reaction time 70 h.
