目的:在荷瘤 BALB/c 小鼠观察热休克小鼠肝癌细胞(H_(22))源 Exosomes(HS-Exo) 激发宿主抗肝癌免疫应答的效应.方法:用自行分离纯化的小鼠肝癌细胞 H_(22) 源 Exosomes 及 HS-Exo 免疫小鼠,然后给小鼠皮下接种 H_(22) 肿瘤细胞,观察肿瘤的生长状况.用MTT法检测小鼠脾细胞增殖情况和细胞毒活性;用免疫组织化学法检测肿瘤组织中CD_4~+、CD_8~+T淋巴细胞浸润情况.BALB/c 小鼠皮下接种 H_(22) 细胞,肿瘤长至可触及大小后,分别注射Exosomes及HS-Exo进行治疗,观察各组肿瘤大小及其小鼠存活期.结果:与对照组 Exosomes 相比,HS-Exo 能使肿瘤出现时间延迟,肿瘤生长缓慢(P<0.05);更能促进脾淋巴细胞增殖并增强其细胞毒活性(P<0.05),能更有效的诱导 CD_4~+ 和 CD_8~+T 细胞活化(P<0.05);更为显著的抑制了小鼠的肿瘤生长(P<0.05).结论:HS-Exo 具有更强的免疫活性,能更好的诱导小鼠抗 H_(22) 肝癌作用,有针对亲本细胞的免疫保护作用,并且具有更为显著的肿瘤治疗作用.

목적:재하류 BALB/c 소서관찰열휴극소서간암세포(H_(22))원 Exosomes(HS-Exo) 격발숙주항간암면역응답적효응.방법:용자행분리순화적소서간암세포 H_(22) 원 Exosomes 급 HS-Exo 면역소서,연후급소서피하접충 H_(22) 종류세포,관찰종류적생장상황.용MTT법검측소서비세포증식정황화세포독활성;용면역조직화학법검측종류조직중CD_4~+、CD_8~+T림파세포침윤정황.BALB/c 소서피하접충 H_(22) 세포,종류장지가촉급대소후,분별주사Exosomes급HS-Exo진행치료,관찰각조종류대소급기소서존활기.결과:여대조조 Exosomes 상비,HS-Exo 능사종류출현시간연지,종류생장완만(P<0.05);경능촉진비림파세포증식병증강기세포독활성(P<0.05),능경유효적유도 CD_4~+ 화 CD_8~+T 세포활화(P<0.05);경위현저적억제료소서적종류생장(P<0.05).결론:HS-Exo 구유경강적면역활성,능경호적유도소서항 H_(22) 간암작용,유침대친본세포적면역보호작용,병차구유경위현저적종류치료작용.
Objective:To investigate the induction of antitumor response and its immune mechanism by H_(22) cell-derived heat shocked Exosomes (HS-Exo).Methods:The mice were immunized by H_(22) cell-derived Exosomes and HS-Exo,then inoculated with the wild type H_(22),and finally the tumor growth were observed.Lymphocyte proliferation and specific cytotoxic activity of mice splenic cells was determined by MTT.Infiltration of lymphocytes in tumor tissues was analyzed by immuno-histochemical staining of CD_4~+,CD_8~+.Results:The tumor size was smaller in HS-Exo group (P<0.05).HS-Exo could induce more lymphocyte proliferation (P<0.05).HS-Exo could generate stronger specific cytotoxic spleen lymphocyte response.Obviously increased infiltrations of CD_4~+,CD_8~+ T cells were observed in the tumor sites of HS-Exo group(P<0.05).And HS-Exo was more effective to restrain tumor growth(P<0.05).Conclusion:Vaccination of H_(22) cell-derived HS-Exo is more effective in the induction of protective immunity and could exert better effect on H_(22) to treat the experimental tumor.
