目的 探讨信号转导和转录激活因子3( STAT3)的临床应用价值.方法 利用电化学发光免疫法检测人肺腺癌细胞A549和人胚胎成纤维细胞MRC-5培养3d的培养液中癌胚抗原(CEA)的表达水平.采用荧光定量PCR法检测人肺腺癌细胞A549和人胚胎成纤维细胞MRC-5中STAT3mRNA、CEA mRNA的表达.结果 CEA在A549细胞培养液中的表达水平高于MRC-5细胞培养液[(31.38±30.58) ng/Lvs (3.10±3.70) ng/L,P<0.05];STAT3 mRNA、CEA mRNA在A549细胞中的表达水平(6.96±3.37,5.68±2.70)均明显高于MRC-5细胞(1.01±0.19,1.02±0.21)(P<0.01),且在A549细胞中STAT3和CEA的表达均呈正相关(r=0.521,P=0.047).结论 STAT3有望成为一种新的诊断肺腺癌的肿瘤标志物.

Objective To investigate the clinical significance of signal transducer and activator of trascription 3 (STAT3).Methods The electrochemiluminescence was used to detect the levels of carcinoembryonic antigen (CEA) in medium which cultured lung adenocarcinoma cell A549,human normal lung cell MRC-5 3 days.Relative expression of mRNA of STAT3,CEA in lung adenocarcinoma cell A549 and human normal lung cell MRC-5 was tested by real time PCR.Results The expression of CEA in lung adenocarcinoma cell A549's medium was higher than that in human normal lung cell MRC-5 [(31.38±30.58) ng/L vs (3.10±3.70) ng/L,P <0.05].The mRNA levels of STAT3,CEA in A549 cell (6.96±3.37,5.68±2.70) were apparently higher than those in MRC-5 cell (1.01 ± 0.19,1.02±0.21) (P <0.01). The mRNA levels of STAT3 had a positive correlation with CEA in lung adenocarcinoma cell A549 ( r =0.521,P =0.047).Conclusions STAT3 can be served as a new marker of lung adenocarcinoma.
