目的 探讨在自然衰老过程中血浆自身抗Aβ抗体的发生规律和对AD发病机制的启示.方法 采用一种特殊的组织淀粉样蛋白免疫活性测定方法(TAPIR)检测205例年龄为1~91岁的健康人群血浆免疫染色Tg2576转基因小鼠大脑组织切片中的老年斑核心,随后血浆与人工合成的Aβ多肽(sAβ)、蚁酸可溶性淀粉样Aβ颗粒(FAβ)进行免疫沉降和免疫印迹,以探查血浆自身抗Aβ抗体的免疫特性.结果 自然发生的自身抗Aβ抗体存在于各个年龄段,甚至于儿童和青少年,总发生率为35.6%,其变化规律呈现一种特殊的"U"型年龄相关性曲线.结论 血浆自身抗Aβ抗体的出现先于脑内淀粉样斑块的形成,且与脑内Aβ负荷无关.

Objective To investigate the occurrence rates and changing role of plasma antibodies to Aβs in normal aging and their enlightenment to the pathogenesis of Alzheimer's disease.Methods Paraffin serial sections from Tg2576 mouse brains were immunostained with 205 plasma samples from healthy cases aged from 1 to 91 years old to make a specific tissue amyloid plaque immunoreactivity analysis, and subsequently, immunoprecipitation of plasma with synthetic Aβ peptides and formic acid soluble amyloid Aβ fractions (FAβ) was employed to clarify the immunospecialties of autoantibodies to Aβs. Results Plasma autoantibodies to Aβs were found to be naturally present in all age periods, even in childhood and juvenile, the total occurrence rate was 35.6%, and they showed a special age-related U-typed trend during aging. Conclusions Plasma autoantibodies to Aβ can occur before amyloid plaques in brains, and with no relationship with brain Aβ burden.
