为探讨α辐射诱发体外细胞转化效应与hprt基因突变之间的相关关系。用体外培养的人胚肺细胞, 经0.25-5Gy α粒子照射后,各受照射剂量组细胞半固体琼脂培养集落形成能力明显增加,细胞获得了非锚着依赖性生长能力, 说明人胚肺细胞在α辐射作用下发生了转化细胞hprt基因突变检测。结果表明, 随着α粒子照射剂量的增加, hprt基因变异系数逐渐增加。细胞转化与hprt基因突变相关分析证实,两者之间存在显著的线性正相关关系。研究结果显示, α辐射的致癌效应与hprt基因突变密切相关。
為探討α輻射誘髮體外細胞轉化效應與hprt基因突變之間的相關關繫。用體外培養的人胚肺細胞, 經0.25-5Gy α粒子照射後,各受照射劑量組細胞半固體瓊脂培養集落形成能力明顯增加,細胞穫得瞭非錨著依賴性生長能力, 說明人胚肺細胞在α輻射作用下髮生瞭轉化細胞hprt基因突變檢測。結果錶明, 隨著α粒子照射劑量的增加, hprt基因變異繫數逐漸增加。細胞轉化與hprt基因突變相關分析證實,兩者之間存在顯著的線性正相關關繫。研究結果顯示, α輻射的緻癌效應與hprt基因突變密切相關。

The purpose of this study is to investigate the relationship between cell transformation and hprt gene mutation induced by α irradiation in vitro. The results showed that the colony formation efficiency (CFE) of human embryo lung cells in soft agar increased after irradiation with 0.25-5Gy, which meant inducible cell transformation by α irradiation. Mutation frequency of hprt gene was found increased with the increase of the doses of α irradiation. Further analysis indicated the positive linear correlation between CFE and the variation coefficient of hprt gene. So, the carcinogenesis induced by α particles maybe is relative to hprt gene mutation.
