目的:研究紫杉醇对人粘液表皮样癌细胞系MEC-1的细胞毒作用,为临床应用选择最佳给药方案提供依据。方法:应用细胞计数法研究不同药物浓度及不同作用时间细胞毒作用的特点。结果:紫杉醇对MEC-1的最大抑制率为50%~90%,当药物浓度为10 nmol/L时抑制率已接近最大,增加药物浓度10~100倍并不能增加抑制效应。但延长药物暴露时间大于12 h能明显增强抑制效应。即使作用72 h时,10~1 000 nmol/L的抑制率分别为64% 、58%、56%、44%和49%。结论:紫杉醇对人粘液表皮样癌细胞系MEC-1有明显的细胞毒作用,能明显抑制其生长,但抑制作用并不完全。此细胞毒作用的时间依赖性明显大于剂量依赖性。提示临床应用应以延长药物作用时间为目的,以期最大程度上发挥紫杉醇的抗肿瘤效应。

Objective: To determine the sensitivity of human mucoepidermoid carcinoma MEC-1 cells to taxol.Methods:MEC-1 cells were exposed to taxol at the doses of 1,10,100 and 1 000 nmol/L;the alive cells were counted in 24,48 and 72 h respectively.In another experiment the cells were exposed to taxol at 10 and 100 nmol/L for 3,6,12,16,36,48 and 72 h respectively,then the drug was removed (except the 72 h exposure-group) and the cells were cultured with fresh medium until 72 h,and the cells were counted.Results:Immedietely after drug-exposure the inhibition effect of taxol against MEC-1 cells nearly achieved maximal(90%) at the concentration of 10 nmol/L,increasing the concentration to 10~100 fold resulted in no additional inhibition ratio in 24~72 h .However when the drug was removed after the cells had been treated at 10 and 100 nmol/L and then cultured for more than 12 hours the inhibition effect was significantly increased.Conclusion:The reaction of MEC-1 cells to Taxol depends more on exposure time than on drug concentration.
