目的 应用血清药理学方法研究中药千里光抗金黄色葡萄球菌的作用及其机制.方法 从千里光中分离出黄酮类化合物,然后通过给小鼠灌胃制备千里光水浸液的含药血清和黄酮类化合物的含药血清.用扫描电镜观察各含药血清对金黄色葡萄球菌超微结构的影响,用同位素前体参入试验观察各含药血清对金黄色葡萄球菌生物合成的影响.同时设立生理盐水血清作为阴性对照.结果 与阴性对照血清相比,两种含药血清作用后的金黄色葡萄球菌,其形态和超微结构均发生明显的变化,菌体塌陷,细菌融合成团,呈现肿胀模糊等变化;其DNA,RNA 蛋白质和肽聚糖合成也均受到明显的抑制.结论 千里光的抗金黄色葡萄球菌作用机制可能是通过抑制细菌的DNA,RNA,蛋白质和肽聚糖的合成有关,其作用的有效成分可能是黄酮类化合物.

목적 응용혈청약이학방법연구중약천리광항금황색포도구균적작용급기궤제.방법 종천리광중분리출황동류화합물,연후통과급소서관위제비천리광수침액적함약혈청화황동류화합물적함약혈청.용소묘전경관찰각함약혈청대금황색포도구균초미결구적영향,용동위소전체삼입시험관찰각함약혈청대금황색포도구균생물합성적영향.동시설립생리염수혈청작위음성대조.결과 여음성대조혈청상비,량충함약혈청작용후적금황색포도구균,기형태화초미결구균발생명현적변화,균체탑함,세균융합성단,정현종창모호등변화;기DNA,RNA 단백질화태취당합성야균수도명현적억제.결론 천리광적항금황색포도구균작용궤제가능시통과억제세균적DNA,RNA,단백질화태취당적합성유관,기작용적유효성분가능시황동류화합물.
Objective To explore the antibacterial mechanism of Senecio scandens Buch-Ham(SSB) on Staphylococcus aureus by serum pharmacological method.Methods Flavonoid compound was isolated from SSB,and then the serum contained water decoct agent of SSB or flavonoid compound were prepared with mice.By scanning electronic microscope (SEM),the effects of drug-containing serum on the ultrastructure of S.aureus were observed.By incorporation experiment of isotope-labeled precursor,the effects of drug containing serum on the biosynthesis of S.aureus were observed as well.Meanwhile,the serum contained normal saline was set as negative control.Results Compared with negative control serum,the morphology and ultrastructure of both drug containing serum on S.aureus showed obvious changes:the cell collapsed,melt together,blurred and swollen,ect.In addition,the synthesis of DNA,RNA,protein and peptidoglycan was inhibited.Conclusion The antibacterial mechanism of SSB on S.aureus may be involved with the inhibition of DNA,RNA,protein and peptidoglycan synthesis,its effective ingredients may be flavonoid compound.
