目的 研究槐定碱对急性心肌缺血大鼠心功能的影响.方法 以结扎左冠状动脉前降支(LAD)方法建立大鼠急性心肌缺血模型,实验分为正常对照组、假手术组、模型组、丹参组、硝酸甘油组、槐定碱2.5,5和10mg/kg剂量组.观察并记录结扎LAD前、结扎LAD后1,5,15,30和60 min的心功能指标的变化.结果 大鼠LAD结扎后, 左心室收缩压(LVSP)、左心室内压最大上升(下降)速率(±dp/dt_(max))明显下降,左心室舒张终未压(LVEDP)明显升高(P<0.01);静脉注射槐定碱2.5,5和10 mg/kg能够改善急性心肌缺血损伤大鼠LVSP 和±dp/dtmax 的变化,降低LVEDP.与假手术组比,各药物组心功能指标降低(除LVEDP升高外),但是与模型组比较均有改善.结论 槐定碱能够改善急性心肌缺血损伤的心功能的变化.

Objective To investigate the effects of sophoridine on the cardiac function in rats with acute myocardial ischemia (AMI) injury. Methods The model was established by ligating the left anterior descending coronary artery (LAD). SD rats were randomly devided into eight groups: normal saline (NS) group, Sham group, AMI group, Danshen (DS) 125 mg/kg group, nitroglycerin (NTG) 0.3 mg/kg group, sophoridine (SR) 2.5, 5, 10 mg/kg groups. To observe and record cardiac function indexs of pre-LAD occlusion,and post-LAD occlusion at 1,5,15,30 and 60 min. The results were compared with those in NS and AMI groups. Results (1) LVSP and ±dp/dtmax were decreased significantly (P<0.01), LVEDP was increased (P<0.01) after LAD occlusion. (2) These results indicated that SR was able to improve LVSP and ±dp/dtmax changes and decrease LVEDP of AMI the dosages of 2.5 mg/kg, 5 mg/kg and 10 mg/kg IV. Compared with sham group, the cardiac function indexes in drug groups were decreased, but they were better than those of NS group, indicating that SR was able to stabilize cardiac function during AMI injury. Conclusion Sophoridine can improve the cardiac function of acute myocardial ischemia injury rats induced by LAD acduion.
