在5L发酵罐下开展毕赤酵母流加培养表达pIFN-α研究.通过多批次研究,结果表明:诱导阶段,RQ稳定情况可以作为反映底物甲醇浓度水平是否合适以及pIFN-α活性高低的监控指标.当甲醇浓度在8~12g/L水平时,RQ在整个诱导阶段能稳定在0.4~0.5之间某个特定值上,同时pIFN-α最高活性都能维持在10~6IU/mL水平上;而当甲醇浓度在0~5g/L或15~20g/L范围内时,RQ很不稳定,分别在0.15~0.4和0.4~0.8区间上下振动,相应的pIFN-α最高活性也只在10~3IU/mL和10~4IU/mL水平上.

Porcine interferon-α (pIFN-α) expression under high density recombinant Pichia pastoris cultivation was conducted in a 5L bench-acaled bioreactor.The results indicated that the stability of respiratory quotient (RQ) could reflect the suitability of methanol concentration for Pichia pastoris induction expression and the pIFN-α activity level indirectly.Maintained with a moderate induction methanol concentration of 8 ~12g/L,RQ could keep constant at a certain value between 0.4 and 0.5 in the whole induction stage.Meanwhile,the highest pIFN-α activity could keep above 10~6 IU/mL.When the methanol concentration was maintained at a lower level of 0 ~ 5g/L or a higher level of 15 ~ 20g/L,the RQ was unstable and the highest pIFN-α activities were at the level of 10~3IU/mL and 10~4 IU/mL,respectively.
