目的 观察携带肝细胞生长因子(HGF)基因的大鼠骨髓间充质干细胞静脉移植是否能够迁移、分布到单侧输尿管梗阻大鼠肾脏.方法 体外分离培养雄性大鼠骨髓间充质干细胞,并转染腺病毒-HGF (Ad-HGF),采用ELISA方法检测转染后HGF的表达情况;建立16只雌性单侧输尿管梗阻大鼠动物模型,随机分为移植组和对照组,移植组经鼠尾静脉注入Ad-HGF转染的骨髓间充质干细胞,对照组注入等量生理盐水,于术后7 d、14d取双侧肾脏,制作石蜡切片,Y染色体原位杂交方法检测雌性受体大鼠.肾脏中是否有骨髓间充质干细胞分布及分布情况.结果 移植组术后7 d梗阻侧肾脏可见Y染色体性别决定基因(Sty)阳性细胞,14d Sry阳性细胞数明显减少(P<0.05),主要分布在肾小管上皮细胞区,肾小球区未见Sry阳性细胞,而梗阻对侧及对照组双侧肾脏未见Sry阳性细胞.结论 鼠尾静脉移植携带HGF基因的骨髓间充质干细胞能够定居于输尿管梗阻侧肾脏,并分布到肾小管上皮细胞区.
目的 觀察攜帶肝細胞生長因子(HGF)基因的大鼠骨髓間充質榦細胞靜脈移植是否能夠遷移、分佈到單側輸尿管梗阻大鼠腎髒.方法 體外分離培養雄性大鼠骨髓間充質榦細胞,併轉染腺病毒-HGF (Ad-HGF),採用ELISA方法檢測轉染後HGF的錶達情況;建立16隻雌性單側輸尿管梗阻大鼠動物模型,隨機分為移植組和對照組,移植組經鼠尾靜脈註入Ad-HGF轉染的骨髓間充質榦細胞,對照組註入等量生理鹽水,于術後7 d、14d取雙側腎髒,製作石蠟切片,Y染色體原位雜交方法檢測雌性受體大鼠.腎髒中是否有骨髓間充質榦細胞分佈及分佈情況.結果 移植組術後7 d梗阻側腎髒可見Y染色體性彆決定基因(Sty)暘性細胞,14d Sry暘性細胞數明顯減少(P<0.05),主要分佈在腎小管上皮細胞區,腎小毬區未見Sry暘性細胞,而梗阻對側及對照組雙側腎髒未見Sry暘性細胞.結論 鼠尾靜脈移植攜帶HGF基因的骨髓間充質榦細胞能夠定居于輸尿管梗阻側腎髒,併分佈到腎小管上皮細胞區.
목적 관찰휴대간세포생장인자(HGF)기인적대서골수간충질간세포정맥이식시부능구천이、분포도단측수뇨관경조대서신장.방법 체외분리배양웅성대서골수간충질간세포,병전염선병독-HGF (Ad-HGF),채용ELISA방법검측전염후HGF적표체정황;건립16지자성단측수뇨관경조대서동물모형,수궤분위이식조화대조조,이식조경서미정맥주입Ad-HGF전염적골수간충질간세포,대조조주입등량생리염수,우술후7 d、14d취쌍측신장,제작석사절편,Y염색체원위잡교방법검측자성수체대서.신장중시부유골수간충질간세포분포급분포정황.결과 이식조술후7 d경조측신장가견Y염색체성별결정기인(Sty)양성세포,14d Sry양성세포수명현감소(P<0.05),주요분포재신소관상피세포구,신소구구미견Sry양성세포,이경조대측급대조조쌍측신장미견Sry양성세포.결론 서미정맥이식휴대HGF기인적골수간충질간세포능구정거우수뇨관경조측신장,병분포도신소관상피세포구.
Objective To assess the migration of hepatocyte growth factor (HGF) transfected bone marrow mesenchyrnal stem cells (MSCS) in murine kidney with unilateral ureteral obstruction (UUO).Methods Bone marrow derived.MSCS from male rats were transfected in vitro with AdHGF.The expression of HGF was measured with ELlSA assay.Sixteen female rats with UUO were randomly divided into two groups:HGF transfected MSCs tansplantation group and control group (saline injection).Kidney tissue of the rats were collected at the end of the 7th day and 14th day postoperatively.The distribution of Y chromosome in the kidney was determined by in situ hybridization.Results Y chromosome positive cells were found only in the obstructed kidneys in the transplantation group.The Y-chromosome positive cells were mainly distributed in the tubular cells.Conclusions MSCs transfected with HGF can immigrate to the rat's kidney with UUO, and are mainly distributed in the region of renal tubular epithelial cells.
