Abrus cantoniensis Hance%Treatment%Explant%Browning rate
[目的]研究影响鸡骨草愈伤组织形成的条件,为鸡骨草的组织培养及悬浮培养提供参考.[方法]从鸡骨草的大田植株和种子发芽后的幼芽中合适部位选取外植体,在附加不同浓度、不同组合的外源激素2,4-D和6-BA的MS培养基上,对鸡骨草愈伤组织进行诱导,计算诱导率,观察愈伤组织生长情况.[结果]单独使用一种激素时对愈伤组织的诱导能力大致相同,2,4-D比6-BA对鸡骨草茎段愈伤组织的诱导能力稍强一些.培养基中添加2种激素时愈伤组织褐变程度稍轻,最适合的培养基为MS附加2,4-D 2.0 mg/L+6-BA 1.5 mg/L.[结论]多种激素组合比单种激素有利于愈伤组织的形成,培养基中适宜的激素组合可降低愈伤组织褐变率.

[Objective] The study aimed to research the condition of influencing the callus formation of Abrus cantoniensis Hance and provide the reference for the tissue culture and suspend culture of A. cantoniensis. [Method] The explants were selected from the suitable position of the plants in the field and the young buds after the seed germination of A. cantoniensis. The calli of A. cantoniensis were induced on the MS media added by the exogenous hormone of 2,4-D and 6-BA with different concn. and different combinations. the inducing rate was calculated and the growth situation of calli was observed. [Result] Using one kinds of hormone singly had a nearly same ability on inducing the calli of A. cantoniensis and 2,4-D showed a slight strong ability on inducing the calli from the stem segment of A. cantoniensis compared with 6-BA. When the medium was added by 2 kinds of hormone, the browning degree of the calli become slight. The most suitable medium was MS medium added by the growth regulators of 2,4-D 2.0 mg/L+6-BA 1.5 mg/L.[Conclusion] Compared with the single hormone, many kinds of hormone combinations were propitious to the callus formation of A. cantoniensis and the suitable hormone combinations added on the medium could reduce the browning rate of the calli.
