[目的]研究不同浓度紫杉醇对A549细胞周期、凋亡和4EBP1分子磷酸化的影响.[方法]采用不同浓度紫杉醇处理A549 细胞,建立凋亡和G2/M期阻滞模型;分别用流式细胞术分析细胞凋亡和细胞周期;用免疫印迹方法分析4EBP1的表达与磷酸化.[结果]G2/M期细胞百分率随紫杉醇浓度的增加而增加,但细胞凋亡率随紫杉醇浓度的增加而下降.随G2/M阻滞的增加,4EBP1分子3组丝/苏氨酸位点磷酸化均增强.[结论]紫杉醇在G2/M期上调A549细胞内4EBP1分子多位点的磷酸化.
[目的]研究不同濃度紫杉醇對A549細胞週期、凋亡和4EBP1分子燐痠化的影響.[方法]採用不同濃度紫杉醇處理A549 細胞,建立凋亡和G2/M期阻滯模型;分彆用流式細胞術分析細胞凋亡和細胞週期;用免疫印跡方法分析4EBP1的錶達與燐痠化.[結果]G2/M期細胞百分率隨紫杉醇濃度的增加而增加,但細胞凋亡率隨紫杉醇濃度的增加而下降.隨G2/M阻滯的增加,4EBP1分子3組絲/囌氨痠位點燐痠化均增彊.[結論]紫杉醇在G2/M期上調A549細胞內4EBP1分子多位點的燐痠化.

[Objective]The paper aimed to study the effects of different concentrations of paclitaxel on the cell cycle and apoptois of A549 and phosphorylation of 4EBP1. [Method]A cell apoptosis and mitosis ( G2/M stage) retention model was established by treating A549 cell with different concentrations of paclitaxel. Cell apoptosis and cell cycle were determined by flow cytometry.The expression and phosphorylation of 4EBP1 were examined by western-blot. [Result]The percentage of mitosis retention of G2/M stage cells raised with the increase of paclitaxel concentrations,but the percentage of apoptosis decreased. The phosphorylation of 3 groups of serine /threonine were all elevated with the mitosis retention.[Conclusion] Paclitaxel enhanced the phosphorylation of 4EBP1 in A549 cellls at G2/M stage.
