从海南洋浦近海分离到1株产植酸酶的酵母菌株,经鉴定为热带假丝酵母(Candida tropicalis),利用Plackett-Burman设计对其发酵条件进行了筛选。结果表明,豌豆粉浓度、硫酸铵浓度和初始pH对酶产量具有显著的影响;利用响应面法对3个因子进行了优化,获得了最佳发酵条件:豌豆粉1.0%,葡萄糖2.0%,硫酸铵1.7%,氯化钠1.0%,初始pH 5.2,培养72 h,植酸酶产量达到727.5 U/mL。

종해남양포근해분리도1주산식산매적효모균주,경감정위열대가사효모(Candida tropicalis),이용Plackett-Burman설계대기발효조건진행료사선。결과표명,완두분농도、류산안농도화초시pH대매산량구유현저적영향;이용향응면법대3개인자진행료우화,획득료최가발효조건:완두분1.0%,포도당2.0%,류산안1.7%,록화납1.0%,초시pH 5.2,배양72 h,식산매산량체도727.5 U/mL。
A phytase producing yeast Candida tropicalis ypy06 was isolated from the coastal water of Yangpu,Hainan.Plackett-Burman design and Response Surface Methodology(RSM) were used to optimize the production conditions of phytase.The results showed that the concentrations of pea,ammonium sulfate and initial pH were the most significant factors to influence phytase production.The optimized composition of medium was 1.0% pea,2.0% glucose,1.7% ammonium sulfate,1.0% NaCl,initial pH 5.2.After optimization,the phytase activity was 727.5 U/mL.
