目的 评价氢气对脓毒症小鼠急性肺损伤的影响.方法 雄性C57BL/6小鼠112只,体重20~25 g,5周龄,采用随机数字表法,将其随机分为4组(n=28):假手术组(A组)、假手术+氢气组(B组)、脓毒症组(C组)和脓毒症+氢气组(D组).采用盲肠结扎穿孔法(CLP)制备脓毒症模型.B组和D组于CLP或假手术后1、6h时分别吸入2%氢气.每组取20只小鼠,记录CLP后7d内生存情况.每组取8只小鼠,于CLP后24 h时处死,采集静脉血样,取肺组织,测定血清及肺组织超氧化物歧化酶(SOD)、过氧化氢酶(CAT)及8-异前列腺素F2α(8-iso-PGF2α)水平,测定支气管肺泡灌洗液(BALF)蛋白浓度和肺组织髓过氧化物酶(MPO)活性,进行肺损伤评分,计算肺湿干重比(W/D).结果 与A组比较,C组生存率、血清和肺组织SOD和CAT活性降低,肺损伤评分、W/D、BALF蛋白浓度、肺组织MPO活性、血清和肺组织8-iso-PGF2α水平升高(P<0.05);与C组比较,D组生存率、血清和肺组织SOD和CAT活性升高,肺损伤评分、W/D、BALF蛋白浓度、肺组织MPO活性、血清和肺组织8-iso-PGF2α水平降低(P<0.05).结论 氢气可减轻脓毒症小鼠急性肺损伤,其机制与抑制氧化应激反应有关.

목적 평개경기대농독증소서급성폐손상적영향.방법 웅성C57BL/6소서112지,체중20~25 g,5주령,채용수궤수자표법,장기수궤분위4조(n=28):가수술조(A조)、가수술+경기조(B조)、농독증조(C조)화농독증+경기조(D조).채용맹장결찰천공법(CLP)제비농독증모형.B조화D조우CLP혹가수술후1、6h시분별흡입2%경기.매조취20지소서,기록CLP후7d내생존정황.매조취8지소서,우CLP후24 h시처사,채집정맥혈양,취폐조직,측정혈청급폐조직초양화물기화매(SOD)、과양화경매(CAT)급8-이전렬선소F2α(8-iso-PGF2α)수평,측정지기관폐포관세액(BALF)단백농도화폐조직수과양화물매(MPO)활성,진행폐손상평분,계산폐습간중비(W/D).결과 여A조비교,C조생존솔、혈청화폐조직SOD화CAT활성강저,폐손상평분、W/D、BALF단백농도、폐조직MPO활성、혈청화폐조직8-iso-PGF2α수평승고(P<0.05);여C조비교,D조생존솔、혈청화폐조직SOD화CAT활성승고,폐손상평분、W/D、BALF단백농도、폐조직MPO활성、혈청화폐조직8-iso-PGF2α수평강저(P<0.05).결론 경기가감경농독증소서급성폐손상,기궤제여억제양화응격반응유관.
Objective To investigate the effect of hydrogen (H2 ) on acute lung injury (ALI) in septic mice.Methods One hundred and twelve male C57BL/6 mice,aged 5 weeks,weighing 20-25 g,were randomly divided into 4 groups ( n =28 each):sham operation group (group A),sham operation + H2 group (group B),sepsis group (group C) and sepsis + H2 group (group D).Sepsis was produced by cecum ligation and puncture (CLP).Groups B and D received 1 h inhalation of 2% H2 at 1 and 6 h after CLP operation or sham operation.Twenty animals in each group were selected and observed for the 7 d survival rate.The left 8 animals in each group were sacrificed at 24 h after CLP operation.Venous blood samples and lung tissues were obtained to determine the levels of superoxide dismutase (SOD),catalase (CAT) and 8-iso-prostaglandin F2alpha (8-iso-PGF2α) in the serum and lungs,the concentration of protein in bronchoalveolar lavage fluid (BALF),and the activity of myeloperoxidase (MPO) in the lungs.The lung injury score (LIS) was assessed and W/D lung weight ratio was calculated.Results Compared with group A,the 7 d survival rate and activities of SOD and CAT in the serum and lungs were significantly decreased,and LIS,W/D ratio,the concentration of protein in BALF,MPO activity and 8-iso-PGF2α level in the serum and lungs were significantly increased in group C ( P < 0.05 ).Compared with group C,the 7 d survival rate and activities of SOD and CAT in the serum and lungs were significantly increased,and LIS,W/D ratio,the concentration of protein in BALF,MPO activity and 8-iso-PGF2α level in the serum and lungs were significantly decreased in group C ( P < 0.05).Conclusion H2 can alleviate ALI in septic mice via inhibiting oxidative stress response.
