以百合新鲜鳞叶为材料,利用RAPD分子标记技术对16份不同生态型的百合种质的遗传多样性和亲缘关系进行了分析.研究结果表明,从120条随机引物中筛选出35条有效引物,共得到769个扩增位点,其中628个位点具有多态性,占81.7%.UPGMA聚类分析结果表明.16份百合种质在阀值为0.9407处分为2个聚类群,即百合科两个不同属的植物:百合属和大百合属;阈值为0.5790处,百合属被分为3个不同的种:百合、卷丹和细叶百合.聚类结果和亲缘关系分析表明不同供试百合之间的遗传多样性较高,亲缘关系较远,且各种质遗传距离与地理距离具有一定的相关性.

In this study, the genetic diversity and phylogenetic relationship among 16 genotypes of Lily collected from different ecological sources was analyzed by RAPD. Thirty-five available RAPD primers were screened from 120 random primers, and a total of 769 DNA bands were amplified, in which 628 bands were polymorphic, the percentage is 81.7%. Cluster analysis showed that the 16 Lily germplasms were divided into two groups at 0.9407. Corresponding to two genus in liliaceae: Lilium and Cardiocrinum, and that germplasms in Lilium were further divided into three different types at 0.5790: Liliam brownii F.E. Brown var.viridulum Baker, L. lancifotium Thunb and L. pumilum DC. Results suggested that the genetic diversity is very high among different Lily germplasms and the genetic distance among germplasms was relative to their geographic distance.
