目的:探讨人宫颈阴道上皮组织中水通道蛋白-2(AQP2)mRNA的表达.方法:采用实时荧光定量PCR技术检测2007年3月至2008年3月首都医科大学附属复兴医院妇产科45例患者(月经正常女性18例,围绝经期女性14例,绝经后期女性13例)宫颈阴道组织AQP2mRNA的表达水平,应用单因素方差分析及一元线性相关与回归进行分析.结果:AQP2mRNA在宫颈阴道上皮中均有表达,月经正常组、围绝经组及绝经组宫颈阴道组织中AQP2mRNA表达量依次降低,月经正常组AQP2mRNA表达水平强于围绝经组和绝经组,差异有统计学意义(P<0.05),围绝经组和绝经组比较,差异无统计学意义(P>0.05),同时研究发现AQP2mRNA表达与年龄呈负相关(r=-0.479,P<0.05),与血浆渗透压呈负相关(r=-0.285,P<0.05),与E2水平呈正相关(r=0.530,P<0.05).结论:围绝经期及绝经后期宫颈阴道上皮中AQP2mRNA的表达下降,可能导致阴道分泌物减少,是引起围绝经期及绝经后期阴道干涩等症的可能机制之一.

목적:탐토인궁경음도상피조직중수통도단백-2(AQP2)mRNA적표체.방법:채용실시형광정량PCR기술검측2007년3월지2008년3월수도의과대학부속복흥의원부산과45례환자(월경정상녀성18례,위절경기녀성14례,절경후기녀성13례)궁경음도조직AQP2mRNA적표체수평,응용단인소방차분석급일원선성상관여회귀진행분석.결과:AQP2mRNA재궁경음도상피중균유표체,월경정상조、위절경조급절경조궁경음도조직중AQP2mRNA표체량의차강저,월경정상조AQP2mRNA표체수평강우위절경조화절경조,차이유통계학의의(P<0.05),위절경조화절경조비교,차이무통계학의의(P>0.05),동시연구발현AQP2mRNA표체여년령정부상관(r=-0.479,P<0.05),여혈장삼투압정부상관(r=-0.285,P<0.05),여E2수평정정상관(r=0.530,P<0.05).결론:위절경기급절경후기궁경음도상피중AQP2mRNA적표체하강,가능도치음도분비물감소,시인기위절경기급절경후기음도간삽등증적가능궤제지일.
Objective:mRNA expression of aquaporin-2 (AQP2) in human ectocervical-vaginal epithelial cell(hECE)was investigated.Methods:45 women were recruit from Capital Medical University Affiliated Fuxing Hospital(18 normal menstruation,14 perimenopause,13 postmenopause),the expression of AQP2 mRNA was detected by Real-time PCR.Univariate analysis and logistic regression were used for analysis.Results:AQP2 mRNA was found in hECE.The expression of AQP2 mRNA was significantly higher in childbearing period than those in peri-menopausal and menopausal periods(P<0.05);while there was no difference between perimenopausal and menopausal group(P>0.05).The expression of AQP2 mRNA and age,plasma osmotic pressure were negatively correlated(P<0.05),while AQP2mRNA and estrogen were positively correlated(P<0.05).Conclusions:Reduced expression of AQP2 mRNA in peri-menopausal and menopausal periods will cause less vaginal secretion,which might be the reason of dryness of vagina.
