目的 研究去甲斑蝥素对肺癌A549细胞株体外放射增敏作用及其可能存在的增敏机制.方法 肺癌A549细胞体外常规培养,通过不同的去甲斑蝥素注射液毒性试验,选取合适的浓度进行增敏试验,分组为空白时照组,单纯加药组,单纯照射组,加药照射组,射线荆量采取2 Gy和5 Gy,采用MTT法测定各组对细胞的抑制率,应用流式细胞仪测定不同浓度药物的诱导细胞周期再分布及其凋亡作用.结果 去甲斑蝥酸钠注射液1 μg/ml组与空白对照组对比,50 μg/ml组与100 μg/ml组对比差异无统计学意义(P>0.05),余各剂量组的OD值之间相比差异有统计学意义(P<0.05).照射药物组与单纯照射组、照射药物组与单纯药物组两两比较差异均有统计学意义(P<0.05).随着去甲斑蝥酸钠注射液浓度的增加,细胞的G2/M比例及凋亡率呈明显增加趋势,经趋势卡方检验,差异有统计学意义(P<0.05).结论 去甲斑蝥素具有放射增敏作用,其放射增敏作用的可能机制为:直接抑制作用、增加对放射敏感的G2/M期细胞比例及诱导凋亡作用.

목적 연구거갑반모소대폐암A549세포주체외방사증민작용급기가능존재적증민궤제.방법 폐암A549세포체외상규배양,통과불동적거갑반모소주사액독성시험,선취합괄적농도진행증민시험,분조위공백시조조,단순가약조,단순조사조,가약조사조,사선형량채취2 Gy화5 Gy,채용MTT법측정각조대세포적억제솔,응용류식세포의측정불동농도약물적유도세포주기재분포급기조망작용.결과 거갑반모산납주사액1 μg/ml조여공백대조조대비,50 μg/ml조여100 μg/ml조대비차이무통계학의의(P>0.05),여각제량조적OD치지간상비차이유통계학의의(P<0.05).조사약물조여단순조사조、조사약물조여단순약물조량량비교차이균유통계학의의(P<0.05).수착거갑반모산납주사액농도적증가,세포적G2/M비례급조망솔정명현증가추세,경추세잡방검험,차이유통계학의의(P<0.05).결론 거갑반모소구유방사증민작용,기방사증민작용적가능궤제위:직접억제작용、증가대방사민감적G2/M기세포비례급유도조망작용.
Objective To investigate the radiosensitizing effect of norcantharidin on lung caner A549 cell line in vitro.Methods Lung cancer A549 cells were conventionally cultured.Select the suitable concentration with different concentrations disodium norcantharidinate injections toxicity test.It was divided into 4 groups:blank control group,drug group,radiation group and drug and radiation group.The dose of ray was 2 Gy and 5 Gy.Detect the OD numerus and calculate the inhibition ratio.Detect the cell cycle and apoptosis which induced by different concentrations disodium noreantharidinate injections with flow cytometry.Results The difference was statistical significance (P < 0.05) between every experiment group and the control group,the differences between radiation drug group and radiation group,radiation drug group and drug group beth were statistical significance (P <0.05).Conclusions Norcantharidinate could induce radiosensitizing effect.Direct repression,increasing the ratio of G2/M and inducing apoptosis could be the possible mechanisms.
