目的 建立人神经母细胞瘤SD幼鼠模型,探讨其作为人神经母细胞瘤非免疫缺陷动物模型用于实验研究的可行性.方法 体外培养人神经母细胞瘤SK-N-SH细胞,取对数生长期瘤细胞,开腹接种于幼年SD大鼠左侧肾上腺区肾筋膜内,接种(1~3)×107/ml的单细胞悬液0.2 ml.定期B超监测肿瘤大小,绘制肿瘤生长曲线,并用免疫组织化学方法检测神经母细胞瘤特异性标志物NSE表达.结果 35只SD大鼠中,荷瘤成功35只,总荷瘤率为100%.其中全身转移有明显恶病质的SD大鼠3只,其余32只为接种局部肿瘤生长.肿瘤生长曲线提示在种瘤后9d左右为肿瘤生长高峰期,种瘤17d左右肿瘤开始缩小.该实验肿瘤生长监测组有2只荷瘤鼠在B超监测1个月左右发生消退,其余荷瘤鼠均在种瘤3个月后消退.所取肿瘤组织免疫组织化学NSE表达均为阳性.结论 成功建立人神经母细胞瘤SD幼鼠模型,该模型荷瘤率高,为研究瘤细胞在非免疫缺陷的正常免疫机体内生长的生物学特性打下良好的基础,为局部瘤体的物理治疗提供经济实用的短期实验动物模型.

Objective To establish neuroblastoma model and explore its feasibility in infancy SD rats. Methods Neuroblastoma cell (Cell line:SK-N SH) was cultured in vitro. (1-3) × 107/ml cells in 0.2 ml of suspension were collected and inoculated into the capsule sac of the left adrenal gland using syringe in SD mouse. The volume of tumor was investigated using ultrasonograph and the growth curve was drawn, furthermore, specific marker NSE of neuroblastoma was investigated by using immunohistochemistry assay. Results Tumor successfully grew in all 35 injected SD rat (100 percent of tumor-formation rate), and 3 cases of cachexia occurred due to metastasis. The tumor tissue grew locally in others rat. The tumor growth curve showed that growth peak occurred on the 9th day after injection and reduced in volume on around 17th day. Tumor tissue in 2 cases regressed in experimental group after 1 month, others regressed after 3 months. Positive NSE expression could be seen in all animals. Conclusions Human neuroblastoma model is available to be established in SD rat with higher formation rate in tumor tissue, which can be utilized to simulate and study their biological characteristics involving their immune mechanism in vivo. It is also ideally experimental model for studying physical therapy of neuroblastoma in economy and practice.
