目的 观察盐酸多奈哌齐(DN)对侧脑室注射Aβ25-35大鼠海马脑源性神经营养因子(BDNF)表达的影响.方法 3月龄Wistar大鼠24只随机分为对照租、干预组、模型组,每组8只,向大鼠侧脑室立体定向注射Aβ25-35建立AD动物模型,然后干预组予以腹腔注射DN,对照组与模型组腹腔注射等量0.9%氯化钠溶液,最后用免疫印迹法检测大鼠海马BDNF的表达.结果 免疫印迹法测定结果显示模型组大鼠海马BDNF的表达量较对照组下降,差异有统计学意义(P<0.01),干预组大鼠BDNF的表达量较模型组增加,差异亦有统计学意义(P<0.05).结论 侧脑室注射Aβ25-35的大鼠其海马BDNF的表达较对照组明显下降,干预组与模型组相比BDNF的表达增加,盐酸多奈哌齐可能会增加AD模型大鼠海马BDNF表达.
目的 觀察鹽痠多奈哌齊(DN)對側腦室註射Aβ25-35大鼠海馬腦源性神經營養因子(BDNF)錶達的影響.方法 3月齡Wistar大鼠24隻隨機分為對照租、榦預組、模型組,每組8隻,嚮大鼠側腦室立體定嚮註射Aβ25-35建立AD動物模型,然後榦預組予以腹腔註射DN,對照組與模型組腹腔註射等量0.9%氯化鈉溶液,最後用免疫印跡法檢測大鼠海馬BDNF的錶達.結果 免疫印跡法測定結果顯示模型組大鼠海馬BDNF的錶達量較對照組下降,差異有統計學意義(P<0.01),榦預組大鼠BDNF的錶達量較模型組增加,差異亦有統計學意義(P<0.05).結論 側腦室註射Aβ25-35的大鼠其海馬BDNF的錶達較對照組明顯下降,榦預組與模型組相比BDNF的錶達增加,鹽痠多奈哌齊可能會增加AD模型大鼠海馬BDNF錶達.
목적 관찰염산다내고제(DN)대측뇌실주사Aβ25-35대서해마뇌원성신경영양인자(BDNF)표체적영향.방법 3월령Wistar대서24지수궤분위대조조、간예조、모형조,매조8지,향대서측뇌실입체정향주사Aβ25-35건립AD동물모형,연후간예조여이복강주사DN,대조조여모형조복강주사등량0.9%록화납용액,최후용면역인적법검측대서해마BDNF적표체.결과 면역인적법측정결과현시모형조대서해마BDNF적표체량교대조조하강,차이유통계학의의(P<0.01),간예조대서BDNF적표체량교모형조증가,차이역유통계학의의(P<0.05).결론 측뇌실주사Aβ25-35적대서기해마BDNF적표체교대조조명현하강,간예조여모형조상비BDNF적표체증가,염산다내고제가능회증가AD모형대서해마BDNF표체.
Objective To explore the effect of donepezil (DN) on the expression of brain-derived neurotrophic factors (BDNF) in hippocampus of rats induced by Aβ25-35.Methods Forty-eight Wistar rats were divided randomly into groups control,DN and model,8 in each.Ad animal models were established by stereotactically injecting Aβ25-35 into the lateral ventricle.DN group were injected with DN for 4 weeks.The expression of BDNF in hippocampus was also determined by Western blotting.Results The expression of BDNF decreased in model group when compared with control group,(P<0.01),and increased in treatment group as compared with model group(P<0.05).Conclusion BDBF expression decreases significantly in rats injected with Aβ25-35,and increases in intervention group as compared with model group.Donepezil hydrochloride may increase BDNF expression in hippocampus of AD model rats.
