细胞因子信号转导抑制因子(SOCS)是一组抑制Janus激酶/信号转导与转录激活因子(JAK/STAT)信号转导的蛋白,由CIS(cytokine inducible SH2 containing protein)、SOCS-1~7组成,在体内町抑制多种细胞因子的信号转导.研究显示,SOCS的一些成员作用于胰岛素受体、胰岛素受体底物(IRS)、磷脂酰肌醇3激酶(PDK)、JAK等胰岛素信号转导分子及瘦素受体(LR)、JAK2、STAT等瘦素相关信号因子,负性调节胰岛素受体信号和瘦素信号的转导,引起胰岛素抵抗和瘦素抵抗,诱发体内葡萄糖代谢失衡,导致糖尿病的发生.

Suppressor of cytokine signaling (SOCS) are the negative regulators of cytokine signaling via inhibition of the Janus kinase/signal transducer and activator of transcription (JAK/STAT) pathway. The family comprises eight structurally homologous members, including CIS (cytokine inducible SH2 containing protein) and SOCS-1 ~7. Current evidence suggests that specific members of the SOCS play a role in the development of leptin and insulin resistance owing to inhibiting leptin and insulin signaling pathways,which results in dysfunction of glucose balance and diabetes. The SOCS proteins inhibit leptin and insulin signaling by influencing multiple proteins, including insulin receptor substrate (IRS), phosphatidylinositol 3-kinase (PI3K) ,JAK leptin receptor (LR),STAT.
