目的 研究凋亡抑制基因Livin在原发性肝癌中的表达,并探讨其与Caspase-3表达的相关性.方法 采用逆转录多聚酶链式反应(RT-PCR)方法检测Livin和Caspase-3在46例原发性肝癌,28例癌旁组织,12例正常肝组织中的表达,分析二者之间相关性.结果 Livin基因在肝癌组织、癌旁组织、正常肝组织表达依次降低,其表达与原发性肝癌患者的年龄、性别、肿瘤大小、是否合并乙肝肝硬化、分化程度、甲胎蛋白等差异均无统计学意义(P>0. 05),而与肝癌是否发生转移有关(P<0. 05);Caspase-3在上述组织中依次升高,差异均有统计学意义,二者在肝癌组织中表达负相关(rs=-0. 333, P<0.05).结论 Livin在肝癌组织中高表达,可能和Caspase-3在原发性肝癌细胞凋亡中起重要作用,可能是原发性肝癌生物学行为和预后重要标记物之一.

목적 연구조망억제기인Livin재원발성간암중적표체,병탐토기여Caspase-3표체적상관성.방법 채용역전록다취매련식반응(RT-PCR)방법검측Livin화Caspase-3재46례원발성간암,28례암방조직,12례정상간조직중적표체,분석이자지간상관성.결과 Livin기인재간암조직、암방조직、정상간조직표체의차강저,기표체여원발성간암환자적년령、성별、종류대소、시부합병을간간경화、분화정도、갑태단백등차이균무통계학의의(P>0. 05),이여간암시부발생전이유관(P<0. 05);Caspase-3재상술조직중의차승고,차이균유통계학의의,이자재간암조직중표체부상관(rs=-0. 333, P<0.05).결론 Livin재간암조직중고표체,가능화Caspase-3재원발성간암세포조망중기중요작용,가능시원발성간암생물학행위화예후중요표기물지일.
Objective To explore the expression of Livin in hepatocellular carcinoma and its relationship with Caspase-3. Methods Expression of Livin and Caspase-3 were detected in hepatocellular carcinoma(n=46), para-carcinoma tissues(n=28) and normal liver tissues(n=12) by reverse transcription polymerase chain reaction (RT-PCR). Results The expression of Livin was found lowered in order in hepatocellular carcinoma, para-carcinoma tissue and normal liver tissue, which had no relationship with age, gender, the sizes of HCC, pathological types and AFP,etc (P>0.05). But it had relationship with the metastasis of HCC(P<0.05).The expression of Caspase-3 was found increased in order in those tissue. Livin expression was negatively related to Caspase-3 expression in hepatocellular carcinoma(rs=-0. 333, P<0.05).Conclusions There is high positive expression rate of Livin in HCC,which may had an important role combined with Caspase-3 in apoptosis of HCC and be a valuable diagnostic and prognostic marker of HCC.
