分离纯化番鸭小肠中碱性磷酸酶,并对其酶促动力学性质进行研究.结果表明:该酶催化磷酸苯二钠水解反应时最适温度为45 ℃,最适pH为9.72,米氏常数(Km)为4.76 mmol·L~(-1).Mg~(2+)对碱性磷酸酶活性有明显激活作用;Cd~(2+)浓度小于0.4 mg·L~(-1)时对碱性磷酸酶有激活作用,而浓度高于0.4 mg·L~(-1)时则有抑制作用;Cu2+对碱性磷酸酶活性有明显抑制作用.
分離純化番鴨小腸中堿性燐痠酶,併對其酶促動力學性質進行研究.結果錶明:該酶催化燐痠苯二鈉水解反應時最適溫度為45 ℃,最適pH為9.72,米氏常數(Km)為4.76 mmol·L~(-1).Mg~(2+)對堿性燐痠酶活性有明顯激活作用;Cd~(2+)濃度小于0.4 mg·L~(-1)時對堿性燐痠酶有激活作用,而濃度高于0.4 mg·L~(-1)時則有抑製作用;Cu2+對堿性燐痠酶活性有明顯抑製作用.
분리순화번압소장중감성린산매,병대기매촉동역학성질진행연구.결과표명:해매최화린산분이납수해반응시최괄온도위45 ℃,최괄pH위9.72,미씨상수(Km)위4.76 mmol·L~(-1).Mg~(2+)대감성린산매활성유명현격활작용;Cd~(2+)농도소우0.4 mg·L~(-1)시대감성린산매유격활작용,이농도고우0.4 mg·L~(-1)시칙유억제작용;Cu2+대감성린산매활성유명현억제작용.
The alkaline phosphatase (AKP) was extracted and purified from the small intestine of Muscovy. The kinetic characters of AKP have been studied. The results showed that the optimum temperature was 45 ℃, and the optimum pH was 9.72 for hydrolysis of penylphosphoric acid disodium salt, the Michaelis constant (Km) was 4.76 mmol·L~(-1). Mg~(2+) can activate AKP significantly, Cd~(2+) can activate AKP at lower concentration than 0.4 mg·L~(-1), but inhibit AKP at higher concentration than 0.4 mg·L~(-1). Cu2+ inhibited AKP significantly.
