目的 观察含母牛分枝杆菌菌苗(微卡)凝胶注入肺部后的吸收和安全性,并对注入的适宜剂量和不良反应进行分析,为可用于肺结核局部免疫治疗提供依据.方法 50只日本长耳兔随机分成5组,每组10只,气管插管至末梢支气管,注入相关制剂.第1组为对照组,注人生理盐水3 ml;第2组为单纯凝胶组,注入卡波姆凝胶3 ml;第3、4、5组为微卡凝胶组,分别注入含3.75、7.50、9.37 mg/L母牛分枝杆菌菌苗的卡波姆凝胶3 ml.持续观察4周,期间检测体温、体质量、血常规,实验结束时处死动物,解剖相关器官及进行病理学观察.结果 各组间体温、体质量、血常规的比较差异均无统计学意义.第2、3组与第1组相比,肺大体及光镜病理观察均未产生明显变化.第4、5组与第1组相比,细支气管未发生明显变化,肺泡结构基本保持完整,间隔轻度增宽,间隔内可见淋巴细胞浸润.结论 含母牛分枝杆菌菌苗凝胶肺内应用具有较好的安全性.
目的 觀察含母牛分枝桿菌菌苗(微卡)凝膠註入肺部後的吸收和安全性,併對註入的適宜劑量和不良反應進行分析,為可用于肺結覈跼部免疫治療提供依據.方法 50隻日本長耳兔隨機分成5組,每組10隻,氣管插管至末梢支氣管,註入相關製劑.第1組為對照組,註人生理鹽水3 ml;第2組為單純凝膠組,註入卡波姆凝膠3 ml;第3、4、5組為微卡凝膠組,分彆註入含3.75、7.50、9.37 mg/L母牛分枝桿菌菌苗的卡波姆凝膠3 ml.持續觀察4週,期間檢測體溫、體質量、血常規,實驗結束時處死動物,解剖相關器官及進行病理學觀察.結果 各組間體溫、體質量、血常規的比較差異均無統計學意義.第2、3組與第1組相比,肺大體及光鏡病理觀察均未產生明顯變化.第4、5組與第1組相比,細支氣管未髮生明顯變化,肺泡結構基本保持完整,間隔輕度增寬,間隔內可見淋巴細胞浸潤.結論 含母牛分枝桿菌菌苗凝膠肺內應用具有較好的安全性.

Objective To provide basis for the local immune therapy of tuberculosis by observing the absorption and security of Mycobacterium vaccae (micro card) gel after administered into lung and analysing the optimum dose and side effects. Methods Fifty Japanese long-eared rabbits were randomly divided into five groups. Group 1 was injected with 3 ml normal sodium. Group 2 was injected with 3 ml carbomer gel. Group 3, 4, 5 were injected with 3 ml carbomer gel with 3.75, 7.50, 9.37 mg/L Mycobacterium vaccae. The temperature, weight, blood were tested durirg four weeks. The animals were sacrificed at the end and tissue pathology was observed. Results There was no statistical significance on temperature, weight, blood among all groups. Compared with group 1, the changes of pathology in lung of group 2 and group 3 were not obvious. There were no obvious changes on the bronchiole between group 4,5 and group 1, alveolar structure remained intact, interval mildly widened,and there was lymphocytic infiltration in interval. Conclusions Gel with Mycobacterium vaccae administered in lung has better security.
