目的 阐明秋水仙碱治疗对血吸虫病肝纤维化小鼠肝脏胶原蛋白表达的影响.方法 应用免疫组化技术,观察血吸虫病肝纤维化小鼠在秋水仙碱治疗前后肝脏Ⅰ、Ⅲ、Ⅵ型胶原蛋白的分布及含量(半定量)变化.结果 秋水仙碱治疗组小鼠与对照组相比,肝内Ⅰ型胶原蛋白无显著变化,而肝内Ⅲ型和Ⅵ型胶原蛋白的免疫组化染色均明显减弱;对胶原含量半定量分级亦显示两组Ⅰ型胶原含量无差异(x2=2.43,P>0.05),而Ⅲ型和Ⅵ型胶原蛋白在治疗组的含量则显著下降(X2分别为6.07,8.17,P<0.05).结论 秋水仙碱治疗可显著降低血吸虫感染小鼠肝内Ⅲ型和Ⅵ型胶原蛋白含量,从而发挥其抗肝纤维化作用.
目的 闡明鞦水仙堿治療對血吸蟲病肝纖維化小鼠肝髒膠原蛋白錶達的影響.方法 應用免疫組化技術,觀察血吸蟲病肝纖維化小鼠在鞦水仙堿治療前後肝髒Ⅰ、Ⅲ、Ⅵ型膠原蛋白的分佈及含量(半定量)變化.結果 鞦水仙堿治療組小鼠與對照組相比,肝內Ⅰ型膠原蛋白無顯著變化,而肝內Ⅲ型和Ⅵ型膠原蛋白的免疫組化染色均明顯減弱;對膠原含量半定量分級亦顯示兩組Ⅰ型膠原含量無差異(x2=2.43,P>0.05),而Ⅲ型和Ⅵ型膠原蛋白在治療組的含量則顯著下降(X2分彆為6.07,8.17,P<0.05).結論 鞦水仙堿治療可顯著降低血吸蟲感染小鼠肝內Ⅲ型和Ⅵ型膠原蛋白含量,從而髮揮其抗肝纖維化作用.
목적 천명추수선감치료대혈흡충병간섬유화소서간장효원단백표체적영향.방법 응용면역조화기술,관찰혈흡충병간섬유화소서재추수선감치료전후간장Ⅰ、Ⅲ、Ⅵ형효원단백적분포급함량(반정량)변화.결과 추수선감치료조소서여대조조상비,간내Ⅰ형효원단백무현저변화,이간내Ⅲ형화Ⅵ형효원단백적면역조화염색균명현감약;대효원함량반정량분급역현시량조Ⅰ형효원함량무차이(x2=2.43,P>0.05),이Ⅲ형화Ⅵ형효원단백재치료조적함량칙현저하강(X2분별위6.07,8.17,P<0.05).결론 추수선감치료가현저강저혈흡충감염소서간내Ⅲ형화Ⅵ형효원단백함량,종이발휘기항간섬유화작용.
Objective To explore the effects of colchicine on the expression of hepatic collagen proteins in schistosomiasis mice with liver fibrosis.Methods Using immunohistochemical technique to observe the changes of the distribution and content of hepatic type Ⅰ、Ⅲ、Ⅳ collagen proteins in schistosomiasis mice with liver fibrosis before and after eolchicine treatment.Results Comparing the colchicine treated group with the control group,the change of hepatic type Ⅰ collagen protein was not significant,but the immunohistochemical staining of hepatic type Ⅲ and Ⅳ collagen proteins was obviously weakened.The type Ⅲ collagen was changed from cord form into wirelike,and the type Ⅳ collagen was also changed from dense reticular form into sparse one.Conclusion Colchicine treatment could reduce the content of hepatic type Ⅲ and Ⅵ collagen proteins significantly in schistosomiasis mice with liver fibrosis,thereby,play its role against hepatic fibrosis.
