目的克隆肾癌相关新基因GYLZ-RCC18的全长,并探讨其功能.方法应用SMART RACE技术克隆GYLZ-RCC18的全长,应用逆转录PCR的方法检测其在肾癌组织和细胞系中的表达特点,以及与肾癌组织分级、分期的关系,通过转染GYLZ-RCC18的反义寡核苷酸阻断其表达,观察其对肾癌细胞系生命特征的变化.结果 GYLZ-RCC18全长约3.5 kb(基因号:BE825133).GYLZ-RCC18在肾癌特异高表达,正常肾表达量极低甚至不表达,两者表达量差别1-9倍之间;GYLZ-RCC18在高分级、高分期肾癌中表达明显高于低分级、低分期肾癌的表达.转染反义基因可明显抑制GRC-1的生长、降低其增殖活性、减少核分裂、并诱导其持续性凋亡.结论本研究表明GYLZ-RCC18是肾癌发生发展密切相关的重要新的癌基因,它的表达与肾癌细胞的生存、增殖、抗凋亡发展密切相关,它的成功克隆为今后肾癌分子机制研究提供了新的理论依据.
目的剋隆腎癌相關新基因GYLZ-RCC18的全長,併探討其功能.方法應用SMART RACE技術剋隆GYLZ-RCC18的全長,應用逆轉錄PCR的方法檢測其在腎癌組織和細胞繫中的錶達特點,以及與腎癌組織分級、分期的關繫,通過轉染GYLZ-RCC18的反義寡覈苷痠阻斷其錶達,觀察其對腎癌細胞繫生命特徵的變化.結果 GYLZ-RCC18全長約3.5 kb(基因號:BE825133).GYLZ-RCC18在腎癌特異高錶達,正常腎錶達量極低甚至不錶達,兩者錶達量差彆1-9倍之間;GYLZ-RCC18在高分級、高分期腎癌中錶達明顯高于低分級、低分期腎癌的錶達.轉染反義基因可明顯抑製GRC-1的生長、降低其增殖活性、減少覈分裂、併誘導其持續性凋亡.結論本研究錶明GYLZ-RCC18是腎癌髮生髮展密切相關的重要新的癌基因,它的錶達與腎癌細胞的生存、增殖、抗凋亡髮展密切相關,它的成功剋隆為今後腎癌分子機製研究提供瞭新的理論依據.

Objective To clone the full length of renal cell carcinoma (RCC) related novel gene GYLZ -RCC18 and study its function.Methods SMART RACE technology was used to clone the full length of GYLZ-RCC18. RT-PCR was used to detect its expression in renal cell carcinoma tissue at different s tages and grades. We transfected the antisense oligonucleotide of GYLZ-RCC18 t o renal cell carcinoma cell line, GRC-1, and analyzed proliferation activity, g rowth rate, apoptosis, and mortality changes.Results The full length of GYLZ-RCC18 (GenBank accession number: BE825133) cDNA was abo ut 3.5kb. GYLZ-RCC18 had a higher expression in higher grades and stage s of renal cell carcinoma than in lower ones. The expression of GYLZ-RCC18 in rena l cell carcinoma was much higher than in normal kidney. After the transfection of GYLZ-RCC18 antisense oligonucleotide, the mortality of GRC-1 increased sig nificantly, while proliferative activity and growth rate were substantially inhi bited at the same time. The antisense oligonucleotide induced apoptosis of GRC -1 through the entire observation time.Conclusion GYLZ-RCC18 is an important novel gene related to renal cell carcinoma. Overexp ression of this gene results in higher growth and proliferative activity and has an antiapoptosis effect on renal cell carcinoma cells. Transfection of the ant isense oligonucleotide may inhibit the generation and development of renal cell carcinoma.
