在摇瓶发酵条件下,采用全因子实验设计法和正交实验设计法对工程菌Hrp-菌株的培养基碳、氮源配方进行优化,确定了最适碳源为葡萄糖、细玉米面,最适氮源为硫酸铵、细豆饼粉;对这四个碳氮源因子的浓度进行L9(34)的正交试验,最终确定出该菌的最适发酵培养基碳、氮源配方为(g/L):葡萄糖8g、细玉米面5g、细豆饼粉35g、硫酸铵2g。优化后的培养基配方使摇瓶发酵液菌量从1.34×10^10cfu/ml提高至3.30×10^10cfu/ml。
在搖瓶髮酵條件下,採用全因子實驗設計法和正交實驗設計法對工程菌Hrp-菌株的培養基碳、氮源配方進行優化,確定瞭最適碳源為葡萄糖、細玉米麵,最適氮源為硫痠銨、細豆餅粉;對這四箇碳氮源因子的濃度進行L9(34)的正交試驗,最終確定齣該菌的最適髮酵培養基碳、氮源配方為(g/L):葡萄糖8g、細玉米麵5g、細豆餅粉35g、硫痠銨2g。優化後的培養基配方使搖瓶髮酵液菌量從1.34×10^10cfu/ml提高至3.30×10^10cfu/ml。
재요병발효조건하,채용전인자실험설계법화정교실험설계법대공정균Hrp-균주적배양기탄、담원배방진행우화,학정료최괄탄원위포도당、세옥미면,최괄담원위류산안、세두병분;대저사개탄담원인자적농도진행L9(34)적정교시험,최종학정출해균적최괄발효배양기탄、담원배방위(g/L):포도당8g、세옥미면5g、세두병분35g、류산안2g。우화후적배양기배방사요병발효액균량종1.34×10^10cfu/ml제고지3.30×10^10cfu/ml。
Optimization of carbon and nitrogen sources for fermentation of Hrp-mutant of Ralstonia solanacearum was conducted applying full factorial design and orthogonal analysis under the condition of flask fermentation.The best carbon sources were glucose and fine corn flour.The best nitrogen sources were(NH4)2SO4 and bean cake powder.Based on the orthogonal test,the best formulation of carbon and nitrogen sources was gained,which was glucose 8 g,fine corn flour 5 g,bean cake powder 35 g and(NH4)2SO4 2 g in 1 L medium.The bacterial biomass was increased from 1.34×1010 cfu /ml to 3.30×1010 cfu /ml.
