以新鲜缢蛏的碎肉为材料,加水配制成一定比例的匀浆,利用超声波提取法对缢蛏碎肉匀浆进行多糖的提取.利用蒽酮-硫酸法对多糖得率进行计算.将得到的多糖粗品通过DEAE离子交换层析及G-75凝胶层析进一步分离纯化.通过邻苯三酚自氧化体系和Fenton体系测定所得样品的抗氧化能力.结果表明:利用超声波提取多糖的提取率为19.93%.多糖粗品通过DEAE离子交换柱,得到2个峰,峰1对超氧阴离子的清除能率为43.89%,质量浓度为 1 mg/ml时,对·OH清除率为 57.80%.峰2对超氧阴离子的清除能率为57.80%,质量浓度为 1 mg/ml时,对·OH清除率为 77.83%.将DEAE峰2进行G-75凝胶层析得到2个峰,峰1对超氧阴离子的清除能率为83.08%,质量浓度为1 mg/ml时,对·OH清除率为94.76%.峰2对超氧阴离子的清除能率为74.41%,质量浓度为1 mg/ml时,对·OH清除率为52.41%.
以新鮮縊蟶的碎肉為材料,加水配製成一定比例的勻漿,利用超聲波提取法對縊蟶碎肉勻漿進行多糖的提取.利用蒽酮-硫痠法對多糖得率進行計算.將得到的多糖粗品通過DEAE離子交換層析及G-75凝膠層析進一步分離純化.通過鄰苯三酚自氧化體繫和Fenton體繫測定所得樣品的抗氧化能力.結果錶明:利用超聲波提取多糖的提取率為19.93%.多糖粗品通過DEAE離子交換柱,得到2箇峰,峰1對超氧陰離子的清除能率為43.89%,質量濃度為 1 mg/ml時,對·OH清除率為 57.80%.峰2對超氧陰離子的清除能率為57.80%,質量濃度為 1 mg/ml時,對·OH清除率為 77.83%.將DEAE峰2進行G-75凝膠層析得到2箇峰,峰1對超氧陰離子的清除能率為83.08%,質量濃度為1 mg/ml時,對·OH清除率為94.76%.峰2對超氧陰離子的清除能率為74.41%,質量濃度為1 mg/ml時,對·OH清除率為52.41%.

The crude product of polysaccharides was isolated from fresh razor clam Sinonovacula constricta chopper added water by ultrasonic extraction and further purified by DEAE ion-exchange chromatography and Sephadex G-75 chromatography. The antioxidant activity of the polysaccharide substances was determined by fenton system and Pyrogallol oxidation. The results showed that the polysaccharide rate of ultrasonic extraction was 19.93%. The razor clam polysaccharides had effect on scavenging hydroxyl free radicals,and the scavenging hydroxyl free radical ability was increased up to 94.76% and scavenging superoxide anion radical ability was up to 83.08%.
