目的:观察大肠腺瘤到癌的演变过程中DNA水平的变化。方法: 应用激光扫描细胞技术(LSC),并采用一种改良的方法,从福尔马林固定石蜡包埋的肿瘤组织中分离细胞,进行细胞分析,特别对伴中到重度不典型增生的病变作深入探讨。结果:发现大肠粘膜上皮随细胞形态学1级到5级的变化,DNA分析图中异倍体的数量也逐渐增加,其中3级和4级(即癌前病变),及5级癌变的病例中异常DI峰数明显增多,后者达5个以上。结论:从该研究可以看到LSC技术能结合细胞形态辩认肿瘤中的异倍体的细小的DI峰,其对大肠肿瘤DNA的分析结果表明异常体的出现数与大肠肿瘤的生物学行为即良恶性有关,在今后的辅助诊断中有一定应用价值。

목적:관찰대장선류도암적연변과정중DNA수평적변화。방법: 응용격광소묘세포기술(LSC),병채용일충개량적방법,종복이마림고정석사포매적종류조직중분리세포,진행세포분석,특별대반중도중도불전형증생적병변작심입탐토。결과:발현대장점막상피수세포형태학1급도5급적변화,DNA분석도중이배체적수량야축점증가,기중3급화4급(즉암전병변),급5급암변적병례중이상DI봉수명현증다,후자체5개이상。결론:종해연구가이간도LSC기술능결합세포형태변인종류중적이배체적세소적DI봉,기대대장종류DNA적분석결과표명이상체적출현수여대장종류적생물학행위즉량악성유관,재금후적보조진단중유일정응용개치。
Purpose To study the relationship of the DNA ploidy to the cytologic morphology in the adenoma-carcinoma sequence of the large bowel, especially in adenomas with moderate-severe atypical lesions. Method This study has used a modified rapid method to isolate cells from block of formalin-fixed paraffin embedded tissues, which were then analyzed by laser scanning cytometer (LSC) developed recently. Results As the histologic grades of the tumor cells increased, the aneuploid peaks found in the tumor was higher, especially in grade 3 and 4 (i.e. precancerous lesions). In grade 5 cancer cases there were as high as 5 aneuploid peaks. Conclusions LSC can visually discriminate tumor cells from other cells and find several aneuploid peaks in the tumors. Aneuploid peaks were related to the biologic behavior of these tumors. LSC technique is applicable in clinical practice.
