FSH、EGF是参与卵巢功能调节的重要因素。关于EGF对FSH促进的胚胎卵巢卵泡发育和类固醇激素发生的影响报道甚少。本实验以单个卵巢的总卵泡数、生长卵泡数和5个最大卵母细胞的平均直径为卵泡发育优劣的衡量指标,利用我们早期研究建立的无血清培养体系研究了EGF对FSH促进的小鼠胚胎卵巢卵泡发育和分泌雌二醇的作用。结果发现: EGF+FSH+雄烯二酮(A)处理组的胚胎卵巢总卵泡数在培养第7~9 d显著高于FSH+A处理组,而且生长卵泡数和卵巢内5个最大卵母细胞平均直径在培养后期也显著高于FSH+A处理组(P<0.05); FSH+A处理组的胚胎卵巢在培养第7~9 d分泌大量雌二醇,而EGF和FSH联合使用时胚胎卵巢在培养早期雌二醇分泌水平显著低于FSH+A处理组(P<0.05)。结果提示EGF协同FSH促进小鼠胚胎卵巢卵泡体外生长发育;同时EGF抑制FSH诱导的雌二醇分泌,其促卵泡发育作用可能与雌二醇的合成不相关联。

Follicle-stimulating hormone(FSH) and Epidermal Growth Factor(EGF)are important regulatory proteins, which involve the regulation of ovarian function. Very few informations were reported on the regulation of EGF in follicle development and steroidogenesis of fetalovary. The present experiment was conducted to evaluate the regulation of EGF on the FSH-induced follicular development and estradiol secretion of mouse fetal ovary in a serum-free condition. The average number of all oocytes, the growing oocyte per-ovary and the average diameter of the five largest growing oocytes were appiled as the index of follicular development. The results revealed that: The total number of oocyte in EGF+FSH+A group was significantly higher than that in FSH+A group between day 7 to 9 in the culture period (P<0.05). The growing oocyte number and the average diameter of the five largest oocytes in EGF+FSH+A group were significantly prior to that in FSH+A group in thelate culture period (P<0.05). A higher level of estradiol in FSH+Agroup was detected from 7 to 9 culture days, while at the same periodonly low concentration of estradiol existed in EGF+FSH+A group. The results showed that EGF had a synergic role with FSH in follicular development of mouse fetal ovary, but which attenuates the production of estradiol induced by FSH in vitro. This suggested that the stimulatory effect of EGF on follicular development is independent of the estradiol production.
