目的 通过检测不同时期血管瘤组织中碱性成纤维细胞生长因子(bFGF)及其受体(FGFR)的mRNA表达水平,初步探讨bFGF及其受体对血管瘤生长的影响,以进一步探讨血管瘤增生退化的机制.方法 取不同时期血管瘤组织及其邻近组织标本,其中增生期20例,退化期12例,正常皮肤组织8例作为对照.根据NCBI基因库中bFGF和FGFR的cDNA序列设计引物,以β-actin为内参照,应用逆转录聚合酶链式反(RT-PCR)方法半定量测定组织中bFGFmRNA和FGFRmRNA表达水平.并分析FGFRmRNA与bFGFmRNA表达的相关性.结论 琼脂糖凝胶电泳显示增生期血管瘤、退化期血管瘤、正常组织中bFGFmRNA和FGFRmRNA电泳条带亮度均依次降低;bFGFmRNA表达的相对量分别为增生期1.8495±0.0950、退化期1.3569±0.0417、正常组0.7188±0.0271;FGFRmRNA表达的相对量依次分别为增生期1.4858±0.1034、退化期0.9225±0.0532、正常组0.4144±0.0272;3组组织中bFGFmRNA和FGFRmRNA表达的相对量以增生期为最高,正常组最少.两组血管瘤组织与正常组织相比差异有显著统计学意义(P<0.01),增生期与退化期之间比较差异亦有统计学意义(P<0.01).经Pearson检验分析,FGFRmRNA与bFGFmRNA在增生期和退化期血管瘤组织中的表达具有显著正相关关系(r=0.850,P<0.01;r=0.812,P<0.01),在对照组亦呈正相关(r=0.804,P<0.05).结论 bFGF及其受体在增生期血管瘤组织中高水平表达,在退化期中表达减弱,在正常组织中几乎无表达;FGFRmRNA与bFGFmRNA基因表达在3组组织中具有正相关关系,提示bFGF及其受体参与调控血管瘤的发展过程.

Objective To investigate the expressions of basic fibroblast growth factor(bFGF) and its receptor(FGFR) in different phases of hemangiomas,to probe into the role of bFGF and FGFR in the development of hemangiomas,and to explore the mechanism of hemangiomatous proliferation and regression.Methods The fresh tissue from different phases of hemangiomas and theirs adjacent tissues were collected.The specimens including 20 proliferating phase,12 involuting phase hemangiomas,and 8 normal tissues as control group.According to the cDNA sequences of bFGF and FGFR in NCBI Gene Bank,the primers were designed and β-actin was used for internal competitive reference standard.The expressions of bFGF and FGFR mRNAs were examined by reverse transcriptase polymerase chain reaction(RT-PCR).The relation of FGFR mRNA and bFGF mRNA was analyzed by correlation analysis.Results The fluorescence brightness of bFGF mRNA and FGFR mRNA electrophoresis strap in the proliferating hemangioma,involuting hemangioma and normal skin was decreased in order.The relative expression of bFGF mRNA in tissues of proliferative hemangioma and involuting hemangioma was 1.8495±0.0950 and 1.3569±0.0417,which was significantly higher than that of normal skin(0.7188±0.0271).The relative expression of FGFR mRNA in the tissues of proliferative hemangioma,involuting hemangioma and normal skin was 1.4858±0.1034,0.9225±0.0532,and 0.4144±0.0272,respectively.By comparing the relative quantities of bFGF mRNA and FGFR mRNA,the highest expressions were in proliferating phase hemangiomas and the lowest were in normal skin.The relative expression quantities of bFGF mRNA and FGFR mRNA were obviously higher in hemangiomas than those in normal skin(P<0.01).The relative quantities of bFGF mRNA and FGFR mRNA in hemangiomas were also higher at proliferative stage than those at degenerative stage(P<0.01).There were significantly positive correlation between FGFR mRNA and bFGF mRNA in the tissue of proliferative stage hemangiomas and involuting stage hemangiomas(r=0.850,P<0.01;r=0.812,P<0.01).And it also showed positive correlation in control group(r=0.804,P<0.05).Conclusions The expressions of bFGF and FGFR are increased in proliferating phase hemangioma,and decreased in involuting phase,but nearly blanked in control group.The positive correlation of FGFR mRNA and bFGF mRNA can be noted among proliferative hemangioma,involuting hemangioma or normal skin.
