A reversed-phase high performance chromatography with evaporative light-scattering detector (ELSD) using methanol-water (4∶1,v/v) as mobile phase was established for direct purity examination of trehalose. An ODS column Zorbax C18 was employed. Regression equations reveal linear relationship between logarithm of trehalose concentration and logarithm of trehalose peak area detected by ELSD in the range of 30~1000 mg/L trehalose. Recoveriy of trehalose measured by ELSD is 99.42% with detection limit of 5 mg/L.
