[目的]初步探讨红酵母原生质体制备和再生的条件.[方法]以红酵母为试材,研究渗透压稳定剂pH值、酶浓度、酶解时间、预处理时间4个因素对红酵母原生质体制备率和再生率的影响.[结果]pH值在6.5~7.0,原生质体的制备率和再生率随着渗透压稳定剂pH值的升高而上升.当酶浓度从0.2%上升到1.0%时,原生质体制备率上升较快,而原生质体再生率下降较慢.随着酶解时间的增加,原生质体的制备率逐渐增加,而原生质体的再生率逐渐下降.在5~15 min,原生质体制备率和再生率均随着预处理的时间的增加而降低.[结论]当渗透压稳定剂pH值为7.0,蜗牛酶酶浓度为1.0%,酶解时间为60 min,预处理时间为5 min时,原生质体的制备率和再生率能分别达到较高的值.

[목적]초보탐토홍효모원생질체제비화재생적조건.[방법]이홍효모위시재,연구삼투압은정제pH치、매농도、매해시간、예처리시간4개인소대홍효모원생질체제비솔화재생솔적영향.[결과]pH치재6.5~7.0,원생질체적제비솔화재생솔수착삼투압은정제pH치적승고이상승.당매농도종0.2%상승도1.0%시,원생질체제비솔상승교쾌,이원생질체재생솔하강교만.수착매해시간적증가,원생질체적제비솔축점증가,이원생질체적재생솔축점하강.재5~15 min,원생질체제비솔화재생솔균수착예처리적시간적증가이강저.[결론]당삼투압은정제pH치위7.0,와우매매농도위1.0%,매해시간위60 min,예처리시간위5 min시,원생질체적제비솔화재생솔능분별체도교고적치.
[Objective] The aim was to preliminarily research the protoplast preparation and regeneration condition of rhodotorula. [Method] With the rhodotorula as the tested material, the effects of four factors including the pH value of osmotic stabilizer, enzyme concn., enzymolysis time and pretreatment time on the preparation and regeneration rate of Rhodotorula protoplast were studied. [Result] The protoplast preparation rate and regeneration rate were raised with the rising of the pH value of osmotic stabilizer when the pH value was in the range of 6.5-7.0. The protoplast preparation rate was raised faster but the protoplast regeneration rate was descended slower when the enzyme concn. was raised from 0.2% to 1.0%. With the rising of the enzymolysis time, the protoplast preparation rate was increased gradually, but the protoplast regeneration rate was decreased gradually. The protoplast preparation rate and regeneration rate all were decreased with the rising of pretreatment time in the range of 5-15 min. [Conclusion] The protoplast preparation rate and regeneration rate were up to the higher value resp. when the pH value of osmotic stabilizer, the enzyme concn. of snail enzyme, enzymolysis time and pretreatment time was 7.0, 1.0%, 60 min, 5 min resp.
