目的 探讨实验性变态反应性脑脊髓炎(EAE)大鼠脑和脊髓L-选择素的表达和意义.方法 大鼠随机分为正常组、福氏完全佐剂对照组(CFA组)、左旋咪唑对照组(LMS组)和模型组;模型组采用豚鼠髓鞘蛋白匀浆和福氏完全佐剂诱发大鼠急性EAE,观察临床表现并评分;HE染色和Loyez氏髓鞘染色观察病理和髓鞘改变;免疫组化法检测脑和脊髓L-选择素表达阳性的血管数.结果 正常组和对照组大鼠均未出现症状,模型组临床症状发生率为100%,模型组L-selectin表达阳性的血管数的数值为(31.86±1.39),较正常组(1.38±0.18)、CFA组(1.50±0.27)和LMS组(7.25±0.59)表达均明显增高(均P<0.05).HE染色见大脑白质及脊髓血管周围有大量炎性细胞浸润;Loyez氏染色见部分脱髓鞘改变.结论 脑和脊髓L-选择索的表达水平增高在大鼠急性EAE中发挥重要作用.
目的 探討實驗性變態反應性腦脊髓炎(EAE)大鼠腦和脊髓L-選擇素的錶達和意義.方法 大鼠隨機分為正常組、福氏完全佐劑對照組(CFA組)、左鏇咪唑對照組(LMS組)和模型組;模型組採用豚鼠髓鞘蛋白勻漿和福氏完全佐劑誘髮大鼠急性EAE,觀察臨床錶現併評分;HE染色和Loyez氏髓鞘染色觀察病理和髓鞘改變;免疫組化法檢測腦和脊髓L-選擇素錶達暘性的血管數.結果 正常組和對照組大鼠均未齣現癥狀,模型組臨床癥狀髮生率為100%,模型組L-selectin錶達暘性的血管數的數值為(31.86±1.39),較正常組(1.38±0.18)、CFA組(1.50±0.27)和LMS組(7.25±0.59)錶達均明顯增高(均P<0.05).HE染色見大腦白質及脊髓血管週圍有大量炎性細胞浸潤;Loyez氏染色見部分脫髓鞘改變.結論 腦和脊髓L-選擇索的錶達水平增高在大鼠急性EAE中髮揮重要作用.
목적 탐토실험성변태반응성뇌척수염(EAE)대서뇌화척수L-선택소적표체화의의.방법 대서수궤분위정상조、복씨완전좌제대조조(CFA조)、좌선미서대조조(LMS조)화모형조;모형조채용돈서수초단백균장화복씨완전좌제유발대서급성EAE,관찰림상표현병평분;HE염색화Loyez씨수초염색관찰병리화수초개변;면역조화법검측뇌화척수L-선택소표체양성적혈관수.결과 정상조화대조조대서균미출현증상,모형조림상증상발생솔위100%,모형조L-selectin표체양성적혈관수적수치위(31.86±1.39),교정상조(1.38±0.18)、CFA조(1.50±0.27)화LMS조(7.25±0.59)표체균명현증고(균P<0.05).HE염색견대뇌백질급척수혈관주위유대량염성세포침윤;Loyez씨염색견부분탈수초개변.결론 뇌화척수L-선택색적표체수평증고재대서급성EAE중발휘중요작용.
Objective To investigate the effect of L-selectin in experimental allergic encephalomyelitis (EAE) of Wistar rats.Methods The rats were randomly divided into four groups;the normal group,the CFA group, the LMS group and the model group;The animal models were established in rats by immunization with myelin basic protein of spinal cord of guinea pig and complete Freund's adjuvant(CFA).The symptom of EAE was observed; pathological feature and myelin of brain and spinal were detected with HE stain and Loyez's stain respectively.The number of positive vessels of L-selectin expression was detected by immunohistochemistry.Results 100% experimental Wistar rats treated with MBP and levamisole developed EAE,but none of the other groups.The number of positive vessels of L-selectin expression was (31.86 ± 1.39) in model group, obviously higher than that of in the normal group (1.38 ±0.18) ,the CFA group( 1.50 ±0.27) and the LMS group(7.25 ±0.59) (all P <0.05) ;The inflammation cells were found around vessels and demyelination were seen in white matter of brain and spinal cords.Conclusion The expression of L-selectin should play an important role in EAE.
