目的 检测大肠癌组织中核不均一性核糖核蛋白(hnRNPA1)和原癌基因(C-myc)的表达,探讨两者在转录调控通路中的相互作用.方法 收集60例大肠癌组织标本,免疫组织化学及半定量聚合酶链反应法(RT-PCR)检测hnRNPA1和C-myc蛋白及基因表达情况.结果 60例大肠癌标本中有35例(58.3%)hnRNPA1蛋白阳性表达,且表达强度随大肠癌分化程度降低而递增(P<0.05);C-myc蛋白共有38例(63.3%)阳性表达;RT-PCR见hnRNPA1阳性标本占96.7%(58/60),C-myc基因阳性标本占91.7%(55/60),两者表达均随大肠癌的分化程度递减而增加(P<0.05);hnRNPA1和C-myc在基因和蛋白质水平呈正相关,r值为0.32和0.42.结论 hnRN-PA1和C-myc在不同分化程度的大肠癌中表达差异有统计学意义,两者在基因和蛋白表达水平上均呈正相关.

Objective To detect heterogeneous nuclear ribonucleoprotein A1 (hnRNPA1) and C-myc mRNA and protein expression in colorectal cancer tissues for further study on mutual action of them in transcription regulation pathway. Methods In 60 coloroctal cancer tissue samples, the expression of hnRNPA1 and C-myc protein and mRNA was detected by using S-P immunohistochemistry and bemi-quan-tiffed reverse transcription polymerase chain reaction (RT-PCR). Results In 60 colorectal cancer tissue samples,35 cases (58.3%) were positive for the expression of hnRNPA1 protein and the expression in-tensity was increased with the decreases in differentiation degrees (P < 0.05) ; 38 cases were positive for the expression of C-mye protein. RT-PCR findings suggested that the positive expression rate of hnRNPA1 mRNA was 96.7% (58/60) and that of C-myc mRNA was 91.7% (55/60) ,and the expression of beth hnRNPAI and C-myc was increased with the decreases in differentiation degrees (P <0.05). There were positive correlations between hnRNPA1 and C-myc mRNA and protein (r = 0.32 and 0.42, respectively).Conclusion There was significant difference in the expression of hnRNPA1 and C-mye in colorectal cancers with different differentiation degrees, hnRNPA1 and C-myc were positively related in gene and pro-tein expression.
