目的 观察Hic-5/雄激素受体相关蛋白55(ARA55)对大肠癌细胞和皮下种植瘤超微结构的影响.方法 在电子显微镜下观察LoVo细胞、稳定转染空质粒和稳定转染Hic-5/ARA55质粒的LoVo细胞的细胞器、细胞连接等超微结构.建立裸鼠皮下种植瘤模型,7周后收取瘤体标本,在电子显微镜下观察各组瘤体的细胞器、细胞连接等超微结构.结果 电子显微镜下显示LoVo-Hic-5/ARA55组细胞和种植瘤体细胞连接数量增多,并且发育良好;胞质中的细胞器较多,主要为粗面内质网.LoVo、LoVo-Voctor组细胞和种植瘤体细胞连接少,且发育不良;胞质中的细胞器少,但游离核糖体丰富.结论 Hic-5/ARA55可使大肠癌细胞的细胞连接数量增加且发育良好,细胞向成熟发展,这可能是Hic-5/ARA55在大肠癌发生、发展中的作用机制之一.

목적 관찰Hic-5/웅격소수체상관단백55(ARA55)대대장암세포화피하충식류초미결구적영향.방법 재전자현미경하관찰LoVo세포、은정전염공질립화은정전염Hic-5/ARA55질립적LoVo세포적세포기、세포련접등초미결구.건립라서피하충식류모형,7주후수취류체표본,재전자현미경하관찰각조류체적세포기、세포련접등초미결구.결과 전자현미경하현시LoVo-Hic-5/ARA55조세포화충식류체세포련접수량증다,병차발육량호;포질중적세포기교다,주요위조면내질망.LoVo、LoVo-Voctor조세포화충식류체세포련접소,차발육불량;포질중적세포기소,단유리핵당체봉부.결론 Hic-5/ARA55가사대장암세포적세포련접수량증가차발육량호,세포향성숙발전,저가능시Hic-5/ARA55재대장암발생、발전중적작용궤제지일.
Objective To investigate the effects of hydrogen peroxide-inducible clone 5 (Hic-5 )/ androgen receptor-associated protein 55 ( ARA55) on the infrastructures of the human colorectal cancer cells and subcutaneous implanting tumors. Methods The ultrastructures of LoVo cells, LoVo cells stably transfected with empty vector (PCDNA4A) and the LoVo cells stably transfected with vector containing Hic-5/ARA55 were observed respectively under an electron microscope. The cells from each group were inoculated subcutaneously into 7 nude mice ( BALB/C nu/nu) respectively. Seven weeks later, the subcutaneous tumors were harvested, and the ultrastructures were observed under the electron microscopy. Results The cell junctions in LoVo-Hic-5/ARA55 cells and subcutaneous tumors were increased and well developed, and organelles were also increased, especially for rough endoplasmic reticulum (RER). On the contrary, cell junctions and organelles were reduced in other two groups, but there were abundant free ribosomes. Conclusion Hic-5/ARA55 could make the cell junctions of LoVo cells increase and well develop, and could make LoVo cells differentiate to mature, which perhaps was one of the mechanisms of the effects of Hic-5/ARA55 on the colorectal cancer.
