目的 探讨色素上皮衍生因子(PEDF)基因在结直肠癌组织中的表达及其与临床病理特征之间的关系.方法 采用荧光定量PCR(FQ-PCR)方法检测59例结直肠癌组织及相应癌旁组织中PEDF mRNA的表达水平.结果 59例结直肠癌组织及相应癌旁组织中均可检测出PEDF mRNA的表达,结直肠癌组织中PEDF的表达量为(5.64±0.66)拷贝数/μg,癌旁组织中PEDF的表达量为(6.88±0.67)拷贝数/μg(P=0.007).早期(Dukes A、B期)结直肠癌患者和肿瘤分化程度高者PEDF的表达量显著高于晚期(Dukes C、D期,P=0.003)和肿瘤分化程度低者(P=0.010).结论 PEDF mRNA的水平与结直肠癌的形成密切相关,初步结果显示PEDF可能是结直肠癌发生、发展过程中的一种保护因素.

Objective To study the expression of pigment epithelium-derived factor PEDF gene in colorectal cancer(CRC) and to investigate the relationship between the expression of PEDF mRNA and the clinic pathologicaI features of CRC.Methods The expression of PEDF mRNA in 59 samples of CRC tissues and matched par cancerous colorectal tissues was detected by FQRT-PCR.Results All the CRC tissues and the matched par cancerous colorectal tissues had expression of PEDF mRNA.The level of PEDF mRNA in CRC was(5.64±0.66)(1g(copies)/μg RNA),and(6.88±0.67)(1g(copies)/μg RNA)in the matched par cancerous colorectal tissues respectively(P=0.007).PEDF expression was significantly related to Dukes stages and the tumor differentiated degree.Conclusion The expression of PEDF mRNA is remark-ably correlated with the occurrence of CRC.The primary results show that PEDF may be a protective factor in the formation and progression of CRC.
