目的 探讨氨溴索对胃内容物吸入性肺损伤大鼠动物模型的抗炎保护作用.方法 54只健康Sprague-Dawley雄性大鼠随机分为3组:对照组、损伤组、氨溴索组.观察各组大鼠动脉血氧分压(PaO2)、肺组织湿重/干重比(W/D)、髓过氧化物酶(myeloperoxidase,MPO)活性变化;ELISA法测定血清肿瘤坏死因子α (TNF-α)含量;Western blot方法检测肺组织前B细胞克隆增强因子(pre-B-cell colony-enhancing factor,PBEF)蛋白表达;光镜下观察肺组织结构变化.结果 与对照组比较,损伤组各时相点PaO2降低(P<0.05),肺组织W/D以及MPO活性增加(P<0.05),血浆中TNF-α含量增高(P<0.05);PBEF蛋白表达均增高(P<0.05);肺组织出现明显病理组织学损伤.与损伤组比较,氨溴索组各时相点PaO2升高(P<0.05),肺组织W/D以及MPO活性降低(P<0.05),TNF-α含量降低(P<0.05);PBEF蛋白表达均下降(P<0.05);肺组织病理学损伤减轻.结论 在大鼠胃内容物吸入性肺损伤模型中,氨溴索可通过抑制肺组织PBEF表达、抑制炎症介质的释放发挥抗炎保护作用.

Objective To investigate the anti-inflammatory roles of ambroxol in rats' gastric aspiration lung injury.Methods Fifty-four Sprague-Dawley rats were randomly divided into three equal groups:control group,iniury group and ambroxol group.Arterial partial pressure of oxygen (PaO2),wetto-dry weight ratio (W/D) of lung,the activity of myeloperoxidase (MPO) were measured.Tumor necrosis factor-α (TNF-α) of serum were examined by enzyme linked immunosorbent assay.Pre-B-cell colony-enhancing factor (PBEF) was examined by Western blot.Lung pathological change was observed by microscope.Results Compared with control group,PaO2 decreased but W/D of lung,the activity of MPO,TNF-α of serum and PBEF expression in injury group increased (all P <0.05).Compared with injury group,PaO2 increased but W/D of lung,the activity of MPO,TNF-α of serum and PBEF expression in ambroxol group decreased (all P <0.05).The microscopic structure of the control group was normal and the pathological changes of ambroxol group were milder than those of injury group.Conclusions Ambroxol has anti-inflammatory effects by restraining the activity of PBEF and the releasing of inflammatory cytokines on gastric aspiration lung injury.
