目的 探讨结直肠癌组织microRNA-145(miR-145)的表达与患者临床症状,尤其是肿瘤远处转移或浸润生长的关系.方法 收集88例手术切除的结直肠癌组织,42例远离癌组织的正常结直肠组织.提取这些标本的总RNA,在使用微小RNA的引物反转录后,用miR-145特异性引物做荧光定量聚合酶链反应(PCR).结果 荧光定量PCR显示:结直肠癌组织miR-145的表达量明显降低,癌组织和正常组织的miR-145相对水平分别为31.65±27.97和189.39±159.30,两者比较差异有统计学意义(P<0.01).miR-145的低表达,与年龄,性别,肿瘤原发病灶的大小,有无淋巴转移明显相关(P>0.05),但与细胞分化程度,远处转移或浸润其他器官是否发生密切相关.高分化(n=25)、中分化(n=34)和低分化(n=29)的癌组织miR-145均值分别为41.96±38.58、29.69±20.43和26.12±23.13,组间差异有统计学意义(P<0.01).有远处转移或浸润其他器官的19个病例miR-145均值仅为14.45±9.98,远低于癌症患者的均值(P<0.01).结论 结直肠癌组织中miR-145的表达低于正常组织,这与癌细胞的分化程度,远处转移或浸润生长密切相关.

Objective To study the expression of microRNA-145 (miR-145) in colorectal cancer tissues and its relationship to tumor cell differentiation, tumor metastasis or invasion. Methods Colorectal cancer tissues were collected from 88 operated patients. Forty-two cases of normal colorectal samples served as controls. Total RNA was extracted from these tissues and reversely transcribed to cDNA by using microRNA specific primers. miR-145 levels were assayed by real-time polymerase chain reaction (PCR).Results miR-145 relative level in colorectal tumor and nomad colorectal tissues was 31.65 ± 27. 97 and 189. 39 ± 159.30 respectively, with the difference being statistically significant between them ( P <0. 01 ).There was no relationship between miR-145 level and patients' age and sex, size of the tumor, lymph node metastasis (P > 0. 05 ). However, the low expression of miR-145 was related to the differentiation of tumor cells. In the cells with higher differentiation (n =25), moderate differentiation (n =34) and lower differentiation (n =29), miR-145 levels were 41.96 ± 38.58, 29. 69 ± 20. 4 and 26. 12 ± 23. 13 respectively (P <0. 01 ). miR-145 was even lower when the tumor had remote metastasis and invasion to neighborhood organs. In 19 patients with the situations, miR-145 levels were only 14. 45 ± 9. 98, significantly lower then the average level of cancer patients (P < 0. 01 ). Conclusion The expression of miR-145 was reduced in colorectal cancer and related to tumor cell differentiation. In the patients with metastasis and invasion the miR-145 expression level was even lower.
