目的 观察白花蛇舌草水提物对CEM细胞增殖影响及其机制.方法 对CEM细胞进行细胞培养.采用台盼蓝拒染法,镜下计数活细胞数,绘制生长曲线,检测白花蛇舌草水提物对CEM细胞的生长抑制作用;琼脂糖凝胶电泳法检测白花蛇舌草水提物对CEM细胞的诱导凋亡作用.结果 白花蛇舌草水提物终浓度0.64,3.2,16μg/ml均显著抑制CEM细胞的生长,且呈剂量和时间依赖性.琼脂糖凝胶电泳结果显示实验组出现明显的DNA梯形凋亡条带,而空白对照组没有出现梯形条带.结论 白花蛇舌草水提物对CEM细胞生长具有显著抑制作用,诱导肿瘤细胞凋亡可能是其主要机制之一.

목적 관찰백화사설초수제물대CEM세포증식영향급기궤제.방법 대CEM세포진행세포배양.채용태반람거염법,경하계수활세포수,회제생장곡선,검측백화사설초수제물대CEM세포적생장억제작용;경지당응효전영법검측백화사설초수제물대CEM세포적유도조망작용.결과 백화사설초수제물종농도0.64,3.2,16μg/ml균현저억제CEM세포적생장,차정제량화시간의뢰성.경지당응효전영결과현시실험조출현명현적DNA제형조망조대,이공백대조조몰유출현제형조대.결론 백화사설초수제물대CEM세포생장구유현저억제작용,유도종류세포조망가능시기주요궤제지일.
Objective To observe the inhibitory function and the mechanisms of the aqueous extract of Hedyotis diffusa Willd on the CEM cells. Methods Culture the CEM cells. Apply trypan blue exclusion to take count of the live cells in the microscope, protract the growth curve and detect the growth inhibitory function of aqueous extract of Hedyotis diffusa Willd on CEM cells. Agarose gel electrophoresis was used to detect the induced apoptosis effects of Hedyotis diffusa Willd aqueous extract on CEM cells.Results The aqueous extract of Hedyotis diffusa Willd inhibited the growth of CEM cells obviously as the medicine concentration were 0.64μg/ml, 3.2μg/ml or 16μg/ml. The aqueous extract of Hedyotis diffusa Willd could induce apoptosis of CEM cells in dose and time-dependent manner. An obvious apoptotic DNA ladder appeared in the experimental group within the agarose gel electrophoresis, while in the control group was on contrary.Conclusion The aqueous extract of Hedyotis diffusa Willd can inhibit the growth of CEM cell strongly. To induce apoptosis is one of its mechanisms.
