目的 探讨高脂饲料对apoE基因敲除(apoE-/-)小鼠心肌组织中内皮型一氧化氮合酶(endothelialnitric-oxide synthase,eNOS)的含量及基因表达的影响.方法 将13只apoE-/-小鼠随机分为两组,即高脂饲料组(n=7)和普通饲料组(n=6).喂养12周后,测血清总胆固醇(total cholesterol,TC)、三酰甘油(triglyceride,TG)、高密度脂蛋白(high density lipoprotein,HDL)和低密度脂蛋白(low density lipoprotein,LDL)含量.采用Real-Time RT-PCR法检测心肌组织中eNOS的mRNA表达水平,以免疫组化方法检测eNOS蛋白的表达.结果 高脂饲料组TC、TG和LDL明显高于普通饲料组.HDL则低于普通饲料组(P<0.01);高脂饲料组心肌组织中eNOS的mRNA和蛋白水平均低于普通饲料组(P<0.01).结论 高脂饲料导致apoE-/-小鼠发生高脂血症,并引起心肌组织中eNOS的mRNA表达的减少和相应蛋白水平的降低.

목적 탐토고지사료대apoE기인고제(apoE-/-)소서심기조직중내피형일양화담합매(endothelialnitric-oxide synthase,eNOS)적함량급기인표체적영향.방법 장13지apoE-/-소서수궤분위량조,즉고지사료조(n=7)화보통사료조(n=6).위양12주후,측혈청총담고순(total cholesterol,TC)、삼선감유(triglyceride,TG)、고밀도지단백(high density lipoprotein,HDL)화저밀도지단백(low density lipoprotein,LDL)함량.채용Real-Time RT-PCR법검측심기조직중eNOS적mRNA표체수평,이면역조화방법검측eNOS단백적표체.결과 고지사료조TC、TG화LDL명현고우보통사료조.HDL칙저우보통사료조(P<0.01);고지사료조심기조직중eNOS적mRNA화단백수평균저우보통사료조(P<0.01).결론 고지사료도치apoE-/-소서발생고지혈증,병인기심기조직중eNOS적mRNA표체적감소화상응단백수평적강저.
Objective To explore the content and gene expression of endothelial nitric-oxide synthase (eNOS) in heart of hyperlipidemia apoE-/- mice. Methods apoE-/- mice were fed with high fat diet as high-fat diet group (n= 7) and normal fat diet as control group (n = 6) for 12 weeks. Serum lipid profiles including total cholesterol (TC), triglyceride (TG), high density lipoprotein (HDL) and low density lipoprotein (LDL) were detected, eNOS mRNA expression in heart were determined by Real-Time RT-PCR, as well as protein expression by immunohistochemistry. Results Levels of serum TC, TG and LDL clearly increased in high fat diet group, while HDL decreased (P< 0. 01). mRNA and protein expression of eNOS in heart reduced in high fat diet group(P<0.01). Conclusions High fat diet may lead to hyperlipidemia in apoE-/- mice, which may down-regulate eNOS expression and decrease eNOS content in heart.
