目的:本工作试图通过观察慢性炎症痛大鼠脊髓背角环氧合酶(COX-2)蛋白表达的变化,探讨脊髓背角COX-2是否与电针镇痛的作用机制有关.方法:在佐剂性关节炎大鼠的模型上,采用行为学和免疫组化技术两种方法,观察电针治疗后不同时相关节炎大鼠的痛敏评分及脊髓背角COX-2蛋白的表达,并与正常组、模型组和药物组加以对照.结果:电针能明显减小关节炎大鼠痛敏评分的绝对值,多次电针具有显著的累积效应;并且随着电针治疗次数的增加,大鼠脊髓背角COX-2免疫阳性细胞数显著下降.结论:电针阳陵泉和昆仑两穴对佐剂性关节炎大鼠有明显的镇痛作用,并同时抑制其脊髓背角COX-2蛋白的表达.
目的:本工作試圖通過觀察慢性炎癥痛大鼠脊髓揹角環氧閤酶(COX-2)蛋白錶達的變化,探討脊髓揹角COX-2是否與電針鎮痛的作用機製有關.方法:在佐劑性關節炎大鼠的模型上,採用行為學和免疫組化技術兩種方法,觀察電針治療後不同時相關節炎大鼠的痛敏評分及脊髓揹角COX-2蛋白的錶達,併與正常組、模型組和藥物組加以對照.結果:電針能明顯減小關節炎大鼠痛敏評分的絕對值,多次電針具有顯著的纍積效應;併且隨著電針治療次數的增加,大鼠脊髓揹角COX-2免疫暘性細胞數顯著下降.結論:電針暘陵泉和昆崙兩穴對佐劑性關節炎大鼠有明顯的鎮痛作用,併同時抑製其脊髓揹角COX-2蛋白的錶達.
목적:본공작시도통과관찰만성염증통대서척수배각배양합매(COX-2)단백표체적변화,탐토척수배각COX-2시부여전침진통적작용궤제유관.방법:재좌제성관절염대서적모형상,채용행위학화면역조화기술량충방법,관찰전침치료후불동시상관절염대서적통민평분급척수배각COX-2단백적표체,병여정상조、모형조화약물조가이대조.결과:전침능명현감소관절염대서통민평분적절대치,다차전침구유현저적루적효응;병차수착전침치료차수적증가,대서척수배각COX-2면역양성세포수현저하강.결론:전침양릉천화곤륜량혈대좌제성관절염대서유명현적진통작용,병동시억제기척수배각COX-2단백적표체.
Objective: The present study was to observe the expression of COX-2 protein in the dorsal horn of the spinal cord in rats with chronic inflammatory pain to investigate whether COX-2 is involved in the mechanism of electroacupuncture (EA) analgesia. Methods: After EA treatment, the pain score and expression of COX-2 protein in the dorsal horn of spinal cord in rats with complete Freund's adjuvant (CFA)-induced arthritis were measured by the behavioral test and immunohistochemistry technique, and compared with those of the normal group, model group and medicine group. Results: EA could decrease the pain score significantly and repeated EA treatments had the remarkable cumulative analgesic effect; and with the increase in EA treatments, the number of COX-2 immunoreactive cells was obviously decreased. Conclusion:EA at acupoints Yanglingquan (GB 34) and Kunlun (BL 60) had remarkable analgesic effect on adjuvant arthritis rats; and in the meantime, could inhibit the expression of COX-2 protein in the dorsal horn of spinal cord.
