目的 研究整合素(integrins)αvβ5对造血细胞功能的影响。方法 用逆转录病毒载体系统将αv、β5基因转染K562细胞，观察αv、β5亚基的表达；用去除血清的方法，观察造血细胞凋亡和分化时αvβ5和αvβ3的变化；用抗αvβ5单抗屏蔽造血细胞上αvβ5时对凋亡和增殖的影响。结果 将含有β5基因的逆转录病毒导入K562细胞，β5亚基没有得到表达；诱导造血细胞分化和凋亡时，αvβ5表达升高，而αvβ3表达下降；用单抗封闭细胞表面的αvβ5，可抑制去血清诱导的细胞凋亡。结论 造血细胞αvβ5的过度表达可抑制细胞增殖并与细胞凋亡有关。

목적 연구정합소(integrins)αvβ5대조혈세포공능적영향。방법 용역전록병독재체계통장αv、β5기인전염K562세포，관찰αv、β5아기적표체；용거제혈청적방법，관찰조혈세포조망화분화시αvβ5화αvβ3적변화；용항αvβ5단항병폐조혈세포상αvβ5시대조망화증식적영향。결과 장함유β5기인적역전록병독도입K562세포，β5아기몰유득도표체；유도조혈세포분화화조망시，αvβ5표체승고，이αvβ3표체하강；용단항봉폐세포표면적αvβ5，가억제거혈청유도적세포조망。결론 조혈세포αvβ5적과도표체가억제세포증식병여세포조망유관。
Objective To investigate the function of αvβ5 integrin inhematopoietic cells. Methods β5 integrin cDNA was expressed in K562 cells through a retroviral vector system. The changes of the αvβ5 and αvβ3 integrins expression in apoptosis and differentiation induced by serum depletion were observed. Result The β5 integrin cDNA failed to express in K562 cells after the introduction of the β5 integrin retrovirus vector pG β5CHT. Imbalanced expression of αvβ3 and αvβ5 integrins occurred during apoptosis and differentiation induced by serum depletion. Treatment of hematopoietic cells with anti-αvβ5 monoclonal antibody inhibited the cells apoptosis. Conclusion Overexpression of the αvβ5 integrin cDNA in hematopoietic cells was associated with the inhibition of cell proliferation and apoptosis.
